The WAGR Network We are the proud parents of a little boy diagnosed shortly after birth with WAGR Syndrome, resulting from a deletion of his 11th chromosome (p12p14). WAGR Syndrome is a very rare disorder. In 1996 when our son was diagnosed with this genetic disorder we were left with more question than answers. Today we are very proud of our network and the group of people with whom we are associated.
The purpose of "Reaching Out" - The WAGR Network is to provide a forum for the exchange of information between families and professionals. This site is for reference only. Please contact your own medical professionals for advice and treatment.
WAGR is an acronym which stands for the following: ( W )ilms' Tumor, ( A )niridia, ( G )enitourinary anomalies/gonadoblastoma and Mental ( R )etardation. A combination of 2 or more of these symptoms must be present for an individual to be diagnosed with WAGR Syndrome. The only feature that has been present in all documented cases of WAGR Syndrome, with only one known exception, is aniridia. Mental retardation, Wilms' Tumor, and/or gonadoblastoma may or may not be present in affected individuals. WAGR is a rare genetic disorder that is thought to affect males more frequently than females.

Nicholas 7 yrs old Our Story Nicholas is our youngest child, he has a sister who is three years his senior. He was delivered by repeat C-Section in September of 1996. My pregnancy was normal so I had no indication that my child was not going to be a perfectly healthy child.
Shortly after birth his doctors informed us that he had undescended testicles. Before leaving the hospital Nicholas was seen by a urologist. The urologist had told us undescended testicles was a common condition in boys. We were still a little scared and concerned. We had hoped his testes would eventually drop down, as they often do in many cases. We made arrangements for a follow up meeting with him 10 days later.
We received a call from our pediatricians office about a week later. We were told that a blood test revealed a problem with one of Nicholas' chromosomes. The doctor did not go into much details. He did recommend that we meet with someone in genetics. We were more scared, confused and in need of some answers. It was at the follow up appointment with urology that everything became much clearer, or much more vague, depending how you look at things.
I took Nicholas to the appointment alone. I was told that Nicholas was born with what doctors call WAGR Syndrome, resulting from a deletion of the 11th chromosome. I was not prepared for what I was about to hear. His doctor spoke about a kidney tumor, a visual impairment, possible retardation and so much more. Nicholas was crying and my heart was breaking inside. I barely had enough strength or composure to drive home from the doctor's office.

It has been estimated that there are perhaps 300 cases (total) worldwide. Very often, the only information immediately available to your child's doctor is a small paragraph in a large textbook. Your child may or may NOT have or develop all of the symptoms of the disorder. Wilm's tumor WilmÂ’s tumor is a solid, malignant (cancerous) tumor of the kidney. WAGR Syndrome is a rare genetic syndrome in which there is a predisposition to several conditions, including certain malignancies, distinctive eye abnormalities, and/or mental retardation. "WAGR" is an acronym for the characteristic abnormalities associated with the syndrome. The acronym stands for (W)ilms' Tumor, the most common form of kidney cancer in children; (A)niridia, partial or complete absence of the colored region of the eye(s) (iris or irides); (G)onadoblastoma, cancer of the cells that form the testes in males or the ovaries in females (gonads); and Mental (R)etardation. A combination of two or more of these conditions must be present for an individual to be diagnosed with WAGR Syndrome. The clinical picture varies, depending upon the combination of associated abnormalities. The only feature that has been present in all documented cases of WAGR Syndrome, with only one known exception, is Aniridia.

Genes, Chromosomal Regions And Developmental Disorders This laboratory has focused on three areas of research - 1) the WAGR (Wilms tumor, aniridia, genitourinary anomalies, and mental retardation) contiguous gene syndrome; 2) identification, by cross-phylum database search, of human genes that define previously unrecognized ancient conserved sequences; and 3) mutation analysis of the RPGR and RP2 genes in X-linked retinitis pigmentosa. C. elegans genome, led this laboratory to seek other human genes that encode previously unrecognized "class marking" ancestral protein sequences by cross-phylum database searches, the second area of research in this laboratory. X-linked (8%) . Two loci for X-linked RP map to the p11.3-11.4 region of the short arm of the X-chromosome: the RP2 locus and the RP3 locus. The RPGR gene, which encodes a protein with homology to the RCC1 guanine nucleotide exchange factor for the GTPase protein Ran, was identified by others as an RP3 gene, and the RP2 gene was found to specify a protein similar to cofactor C involved in b -tubulin folding.
