    In our case, we have three children, 5-year-old twins without PRS and a 2-year-daughter with PRS. No underlying syndrome has been diagnosed, but I'm not counting anything out yet. At this time the 2-year-old is the only child in our extended family to have any sort of problem. But I assume the twins will have some increased risk of having a child with PRS, even though it may not be a substantial risk. I guess after a year or so of obsessing I made the decision not to carry that worry with me on a daily basis. Since my husband and I don't plan to have additional children, the questions doesn't need to be answered immediately. Hopefully by the time the kids are ready for kids we'll all know more about this, though I can't imagine it would cause them to forego parenthood. Sarah Previous Message

This is a page devoted to indexing on-line information related to Stickler's Syndrome. It has not been thoroughly edited for two years, so it and the links in it are somewhat out-of-date. For the latest information, please see the SIP (US) website at http://www.sticklers.org/ or the Stickler Syndrome Support Group (UK) website at http://www.netcomuk.co.uk/~gfmsssg/ Stickler's Syndrome defines a set of closely-related genetic connective tissue disorders caused by a range of collagen gene mutations. It is an autosomal dominant trait, meaning that it is independent of sex and is expressed in a person with a single copy of the diseased gene. It thus has a 50% chance of being passed from parent to each child. Its expression can vary greatly within and among families, and is often recognized after a child is born with some degree of cleft palate or develops cataracts and/or retinal detachments at an early age. Its other expressions can include extreme myopia, hyperextensibile joints and arthritis, a somewhat flat face and small jaw and high frequency hearing loss. There are a wide range of collagen disorders with overlapping symptoms, so a genetic evaluation is required in order to diagnose Stickler's syndrome. Early and regular eye exams by an informed ophthamologist can make effective treatment of myopia, cataracts, and detached retinas possible. Craniofacial, skeletal, auditory, and cardiac evaluations may be considered. Contact sports should probably be avoided.

Connective tissue, as its name suggests, is involved in connecting different parts of our body together. Stickler Syndrome is caused by changes in the connective tissue. What are the features of Stickler Syndrome?
People with Stickler Syndrome may have a cleft palate and a small lower jaw. These are two features that are part of the Pierre Robin Sequence . Of those with Stickler Syndrome, 60% have Pierre Robin sequence. The cleft palate is surgically repaired around 12-24 months of age. The eyes can be affected, resulting in near sightedness. There is also an increased risk of cataracts and retinal detachment. Hearing loss of some degree affects around 80% of people with Stickler Syndrome. If hearing loss is conductive (sound is not conducted from the external ear to the inner ear), it is often possible to improve it by surgery. However, surgery is frequently unable to improve hearing if the hearing loss is sensorineural (the hearing problem is in the inner ear, auditory nerve or auditory centers of the brain). Hearing aids can help hearing in both types of hearing loss.

General Discussion Stickler syndrome refers to a group of disorders of the connective tissue that involves several of the body’s organ systems such as the eye, skeleton, inner ear, and/or the head and face. Connective tissue is made up of a protein known as collagen that develops into the several varieties found in the body. It is the tissue that physically supports many organs in the body and may act like a glue or an elastic band that allows muscles to stretch and contract. Stickler syndrome often affects the connective tissue of the eye, especially in the interior of the eyeball (vitreous humor), and the ends of the bones that make up the joints of the body (epiphysis). Most authorities agree that there are four types of Stickler syndrome, of which three are reasonably well differentiated and a fourth remains not well understood.

Stickler's syndrome
Stickler syndrome is a fairly common autosomal dominantly inherited disease characterized by a high incidence of retinal detachments, joint problems, cataracts, and facial and dental abnormalities. The retinal detachments are often complex and difficult to repair, and are associated with an abnormality of the interface between the retina and the vitreous jelly. This abnormality is known as lattice degeneration and is much more common than the Stickler syndrome itself. Lattice degeneration is thought to play an important role in a large proportion of non-Stickler retinal detachments. It is hoped that by understanding the pathophysiology of Stickler retinal detachments, that we can gain further insight into the pathogenesis of detachments associated with common lattice degeneration. Stickler patients often suffer complicated retinal detachments that require specialized vitrectomy surgery to repair. Here, sulfur hexafluoride gas has been injected into the eye to help reattach the retina. In 1987, Francomano, et al. used chromosome linkage analysis to implicate a collagen gene (2A) in Stickler syndrome. In 1991, Ahmad, et al actually identified a mutation in this collagen gene in a family with Sticklers. In 1992, we identified a second collagen mutation in one of the eight families with Stickler syndrome that we have been studying. We have subsequently identified four additional Stickler mutations.
