Sotos Syndrome is a rare genetic disorder characterized by excessive growth that occurs prior to and after birth (prenatally and postnatally). At birth, affected infants have unusually increased body length that is abnormal in proportion to weight, which may also be above average; in addition, newborns typically demonstrate advanced bone growth, abnormally large hands and/or feet, and characteristic facial features. Abnormally rapid growth continues after birth until approximately four or five years of age, at which time growth may slow to a normal rate. Adult height usually exceeds the 50th percentile. Some affected individuals may reach excessive adult heights; males of 193 cm to 203 cm (6 feet 4 inches to 6 feet 8 inches) and females up to 188 cm (6 feet 2 inches) have been described.

The Sotos Syndrome Support Association (SSSA) was organized in 1988 to provide a support network for families and individuals affected by the syndrome. Sotos syndrome, also known as cerebral gigantism, is a neurological brain-based disorder resulting in physical and mental developmental delays in children. The goals of the SSSA are to provide a social support environment for families and individuals with Sotos syndrome; increase public awareness and education about the disorder; and provide opportunites for professionals working with affected individuals to collect and share data for research. The SSSA has members across the United States, Canada and Great Britain, and sponsors an annual conference.

Resources for Genetic Counselors site updated May 10, 2004 outlines links search Sotos Syndrome Overgrowth syndrome etiology still uncertain although researchers in Japan found a gene that may be the cause of Sotos syndrome in a large number of individuals with classical Sotos syndrome (published paper in 2002) usually sporadic Incidence: Unknown slightly less common than Beckwith-Wiedemann with prevalence of 1 in 13,700 births Diagnosis: Diagnosis is clinical 4 core features (must have at least 3 of the 4 to really make a diagnosis) First 3 features are relative to general population so setting a cutoff is not always clear rapid early growth pre and postnatal advanced bone age developmental delay characteristic facial appearance specific to Sotos syndrome o alters in childhood and adolescence o in newborn period (macrocephaly, high bossed forehead, dolichocephaly, high palate, appearance suggestive of hypertelorism) o face lengthens, jaw becomes more prominent, dolichocephaly (tall narrow skull) and frontal bossing (prominent forehead) become more obvious o delay in growth of hair which is often thin “appearance of receding hairline” o midchildhood downslant to palpebral fissures, wear and discoloration of teeth, tendency for a rosy coloration of cheeks, chin, and nasal tip.
