Rett Syndrome Research Program Description
Rett syndrome is a debilitating neurologic disorder. Children with Rett syndrome appear to develop normally until six to 18 months of age. They then enter a period of regression, losing speech and hand skills they had acquired. Most develop seizures, repetitive hand movements, irregular breathing, and motor-control problems. They can live to adulthood, but most never regain the ability to use their hands or to speak The cause of the disorder is a defect in the MECP2 gene on the X chromosome. Rett Syndrome Research Team: Research support provided by the Rett Syndrome Research Foundation.

Developmental Epigenetics Group Rett Syndrome
Rett Syndrome (RTT, OMIM #312750 ) is caused by mutations in the X-linked methyl-CpG binding protein MeCP2. Rett Syndrome is found almost exclusively in girls. RTT patients appear normal for the first 6-18 months of life, but then begin to lose the ability to speak or use their hands. This regression continues into autistic behaviour, ataxia, repetitive hand movements, and microcephaly, and often involves breathing abnormalities, seizures, eating problems, and growth retardation. RTT is a relatively common disorder, occurring once every 10,000-15000 live female births.

We suspect that most of these mutations result in a non-functional protein. Many missense mutations which occur within the Methyl CpG binding domain (MBD) are known to interfere with the structure of the domain.

It is often misdiagnosed as cerebral palsy or autism. It primarily occurs in females and it can strike all racial and ethnic groups. Development appears to be on target until toddlers reach six to eighteen months of age. Symptoms range from loss of mobility to the inability to retain acquired speech. Her birth will be like any other, and she will appear healthy in all ways. She will coo, smile and make her parents unbelievably happy. But within six to 18 months of her birth, symptoms of Rett syndrome will present themselves, and she will endure countless hardships, and her parents will face boundless challenges. What is Rett Syndrome?
According to Kathy Hunter, founder and president of The International Rett Syndrome Association , Rett syndrome (RS) is a neurological disorder seen almost exclusively in females and found in a variety of racial and ethnic groups worldwide. The child with RS usually shows an early period of apparently normal development until 6 to 18 months of life. From that point on, the child gradually loses communication skills and purposeful use of the hands. Stereotyped hand movements, gait disturbances and slowing of the rate of head growth become apparent. Other problems may include seizures and disorganized breathing patterns that occur when awake. This syndrome is often misdiagnosed as autism, cerebral palsy or non-specific developmental delay. In the case of Connie Coughlin of Bartlett, Tenn., her daughter, Dani, was diagnosed with cerebral palsy. The family believed that for 11 years until she was officially diagnosed with RS.

"We cannot live only for ourselves. A thousand fibers connect us with our fellow men; and along these fibers, as sympathetic threads,
our actions run as causes, and they come back as effects." Herman Melville When we first found out that anything might be "wrong" with Dani, it was like going through the death of someone you love. You literally go through all the stages of grief; denial, anger, depression, grief and acceptance. Not always in that order, but most experience each emotion. "If you learn from your suffering, and really come to understand the lesson you were taught, you might be able to help someone else who’s now in the same phase you may have just completed. Maybe that’s what it’s all about after all." -Anonymous- We were seeing a group of five (5) Pediatricians and they all kept telling us the same thing, "Each child progresses at a different rate." For almost three years I listened to this. You may wonder why we didn't switch doctors..... part was probably denial, I knew something was wrong, but as long as I had 5 "professionals" telling me I was crazy, I could hold on to the last bit of hope I had.

Study Offers New Insight Into Rett Syndrome
Cambridge, Mass. – Rett Syndrome is a major cause of mental retardation in girls. Although researchers have identified the protein involved in the disease, its exact role remains a mystery. Now, a group of researchers from Children's Hospital Boston and Whitehead Institute of Biomedical Research have identified the protein's function, a discovery the scientists say could be the first significant advance in Rett Syndrome research in years.

Researchers suspect that mutations in the protein impair its ability to regulate genes during a critical stage of brain development.
