Abstract. The aim of the study is to investigate the presence of different mycoplasmal species in blood samples from patients with Chronic Fatigue Syndrome and/or Fibromyalgia Syndrome. We previously found that more than 60% of patients with Chronic Fatigue Syndrome/ Fibromyalgia Syndrome had mycoplasmal blood infections, such as M. fermentans . In this study, patients with chronic Fatigue Syndrome/Fibromyalgia syndrome were examined for multiple mycoplasmal infections in their blood. A total of 91 patients diagnosed with Chronic Fatigue Syndrome/ Fibromyalgia Syndrome and with a positive test for any mycoplasmal infection were investigated for the presence of M. fermentans

Abstract: OBJECTIVE: Mycoplasmal infections are associated with several acute and chronic illnesses. Some mycoplasmas can enter a variety of tissues and cells, and cause system-wide or systemic signs and symptoms. METHODS: Patients (14 female, 14 male) diagnosed with rheumatoid arthritis (RA) were investigated for mycoplasmal infections in their blood leucocytes using a forensic polymerase chain reaction (PCR) procedure. Amplification was performed with genus- and species-specific primers, and a specific radiolabelled internal probe was used for Southern hybridization with the PCR product. Patients were investigated for the presence of Mycoplasma spp., and positive cases were further tested for infections with the following species: M. fermentans, M. hominis, M. pneumoniae and M. penetrans. CONCLUSIONS: The results suggest that a high percentage of RA patients have systemic mycoplasmal infections. Systemic mycoplasmal infections may be an important cofactor in the pathogenesis of RA, and their role needs to be explored further

Enzootic pneumonia caused by Mycoplasma hyopneumoniae is a difficult disease to control and has stretched the minds of farmers, veterinarians and academicians for several decades. On its own the mycoplasmal infection can be quite mild causing occasional coughing, depression of growth rate, feed conversion efficiency and lung lesions can be from zero to moderate, affecting about 25% of the lung experimentally. Under today's intensive production systems, the primary infection is frequently invaded by secondary bacterial infections, especially Pasteurella multocida which can lead to bronchopneumonia, acute respiratory signs (difficulty breathing) and even death, with the lung lesions doubling in size upto 50% of the lung. In addition over recent years, with PRRS virus and PMWS (circovirus type 2), the poor pig's lung receives such an overwhelming infection leading to a complete swamping, with 70-80% of the lung with consolidated lesions. Mycoplasmas are immunosuppressive, i.e. they confuse the immune defence system in the lung to stop it destroying them. It takes upto 4 weeks for the lesions to develop; they then consolidate for a further 4 weeks and then progressively heal from 10 weeks, as the pig's own immunity/resistance develops. Normally a respiratory infection (such as the common cold) is cleared within 14 days. This immuno-suppression allows the secondary bacteria and other primary bacteria such as

February 7, 2002 FDA TO HOLD TECHNICAL WORKSHOP ON SWINE MYCOPLASMAL PNEUMONIA The Food and Drug Administration (FDA) is holding a Swine Mycoplasmal Pneumonia Technical Workshop. The topic to be discussed at this public workshop is how to evaluate drug effectiveness against swine mycoplasmal respiratory disease. The public workshop will be held on March 6 and 7, 2002, at the DoubleTree Hotel Kansas City, 1301 Wyandotte St., Kansas City, MO 64105, 816-474-6664. The workshop will be held from 8:30 a.m. to 5:00 p.m. on Wednesday, March 6 and from 7:00 a.m. to 3:30 p.m. on Thursday, March 7. FDA is seeking scientific input from a broad public forum to help the Agency determine an acceptable method, in light of the current state of scientific knowledge, for evaluating drug effectiveness against swine mycoplasmal respiratory disease. Mycoplasma hyopneumoniae is a major pathogen in "porcine respiratory disease complex" (PRDC). PRDC is a significant problem in the swine industry in the United States and abroad. This workshop will provide a necessary forum for leveraging scientific resources, including top experts in swine mycoplasmal pneumonia. The workshop is part of CVM's leveraging initiative aimed at increasing interaction with industry, academia, practitioners, and other government agencies. Registration is required for the meeting, and space is limited. Registration will be on a first come, first served basis, and there is no registration fee.

Raleigh, NC This swine pneumonia is important because it is difficult to raise pigs free from the organisms which cause the disease, it has been reported to occur wherever pigs are raised throughout the world, and it adversely affects feed intake and growth performance. This disease affects many pigs but does not cause many deaths unless severe secondary infections or complicating diseases occur. It has been called many different names including mycoplasmal pneumonia, mycoplasmal induced respiratory disease, enzootic pneumonia, and others. This pneumonia is a disease complex caused by Mycoplasma hyopneumoniae and made worse by Pasteurella multocida. Both of these organisms are very commonly isolated from the lungs of pigs. Pasteurella is easier to isolate and identify in the laboratory. Whenever mycoplasmal pneumonia is discussed in swine herds, the reality is that both mycoplasmal and pasteurella are present and involved in the disease. The economic significance of mycoplasmal and pasteurella pneumonia is difficult to determine exactly. Estimates in the U.S. swine industry including reduced daily weight gain and antibiotics used to treat the disease are estimated to be $300,000,000 annually. Cost of pig deaths are not included in this number. A 1984 study showed that in 315 slaughter examinations of 9500 pigs, 99% of the herd and 70% of the pigs had pneumonia lesions typical of mycoplasmal pneumonia.

The two biggest problems in cell culturing are misidentification or cross-contamination of cell lines and contamination with microorganisms (e.g. bacteria, fungi, yeast, viruses and above all mycoplasmas). In order to exclude misidentified and contaminated cell lines and to ensure the authenticity of the cultures, we make efforts to perform a number of analyses on cell lines:
  • viability and growth tests for the detection of microorganisms (mycoplasmas, other bacteria, fungi, yeast) tests for the detection of viral contamination PCR analysis for the determination of murine species DNA fingerprinting karyotyping and cytogenetic analysis immunofluorescence staining of cellular antigens.

  • In addition to this standard quality and identity control program, cell lines are characterized in detail with regard to various parameters at the DSMZ; for details see Quality Control, Identity Control, and Characterization of Cell Lines . Those tests that were performed on any given cell line are indicated for each cell line in the individual entries. The specific tests to exclude cross-contamination and mycoplasma contamination are described in detail under Cytogenetics DNA Fingerprinting , and Mycoplasma Detection and Elimination , respectively.
