What is Anophthalmia?
Anophthalmia is a medical term which is used to describe the absence of the globe and ocular tissue from the orbit.
The terms anophthalmia and microphthalmia (small eyes) are often used interchangeably since CT Scans or MRI show some remnants of either the globe or surrounding tissue in most cases.
Anophthalmia/Microphthalmia (A/M) may affect one eye with the other eye being normal, or both eyes, resulting in blindness. Anophthalmia is rare, but the exact incidence is unknown. One report from a prospective study of 50,000 newborns found an incidence of microphthalmia of 0.22 per 1,000 live births. In a recent study in England, the prevalence of anophthalmia and microphthalmia was 1.0 per 10,000 births. Anophthalmia can be congenital (present at birth) or acquired later in life.

DISORDERS GLOSSARY Microphthalmia, Anophthalmia, and Coloboma (as defined by MACS The above photos are the left and right eyes of an 8 year old boy with bilateral microphthalmia and coloboma. This boy is blind in his right eye and partially sighted in his left. The coloboma can be clearly seen. There is a very obvious colour difference in the eyes which is fairly common. Below is a photograph of the boy's left retina. The white area is the coloboma and the red is healthy(ish) retina.

Anophthalmia Primary anophthalmia
is a complete absence of eye tissue due to a failure of the part of the brain that forms the eye. I n secondary anophthalmia
the eye starts to develop and for some reason stops, leaving the infant with only residual eye tissue or extremely tiny eyes which can only be seen under close examination. I n degenerative anophthalmia the eye started to form and, for some reason, degenerated. One reason for this occurring could be a lack of blood supply to the eye.

My daughter MacKenna was born with microphthalmus and I was looking in this area last year for some advice on how to deal with: what at the time seemed like such a horrible ordeal: and now realizing that it was just a blessing in disguise. All the E-mail responses I received from people helped a lot. My daughter is now 13 months old and doing great she has a prosthetic eye that is absolutely perfect. If you did not know her you would not she had microphthalmia. My daughter was born almost a year ago with micropthalmia of the right eye and coloboma of the left. She has a prosthetic eye and it looks wonderful! I am just wondering if anyone else out there has had any genetic testing done and, if so, what were the results? Do your children have siblings and, if so, were they also affected with the same condition? We have recently had genetic testing done and it was inconclusive. We only have one child and we have been told that it is possible that one of us carries the "defective" gene that causes this or that we do not at all. At any rate, the doctors have told us that any other children have a to 50% chance of being affected with either micropthalmia or coloboma or both. We have now also been told that, should the retina of the left eye detach, it is possible that it is not correctable by surgery.

Introduction
Mutations in the Mitf gene can produce codominant, recessive, or dominant inheritance depending on which part of the protein is affected. Codominant inheritance is produced when the mutation is in DNA binding domain. In this case, the protein dimerizes normally - with protein from normal or mutant allele, but heterodimers can not bind DNA normally (fig.7). With recessive inheritance, the mutations are in the dimerization domains. The mutant protein can not dimerize and therefore cannot bind DNA efficiently (fig. 8). Dominant inheritance patterns have only been found in humans. Here, the DNA binding domain is affected. It has been suggested that human development is more sensitive to haploinsufficiency.
Materials and Methods
Single Strand Conformational Polymorphisms (SSCP) and Heteroduplex (HD) analysis of amplified gene segments can effectively screen for single base mutations in nucleic acids. SSCP is based on the fact that single stranded (denatured) DNA molecules take on a sequence specific secondary structure (conformer) when transferred to non-denaturing conditions. Under the same conditions, some of the molecules will renature as a double helix. If there were two DNA sequences, differing only by a single base, heteroduplexes (double helices consisting of one of each sequence) as well as homoduplexes will form. The different conformers of single strands and the different duplexes can be detected due to differing mobilities in gel electrophoresis. used to identify linkage or homology of the merle gene with the mouse or human mitf gene.
