WHAT IS KLINEFELTER SYNDROME? In 1942, Dr. Harry Klinefelter and his coworkers at the Massachusetts General Hospital in Boston published a report about nine men who had enlarged breasts, sparse facial and body hair, small testes, and an inability to produce sperm. By the late 1950s, researchers discovered that men with Klinefelter syndrome, as this group of symptoms came to be called, had an extra sex chromosome, XXY instead of the usual male arrangement, XY. (For a more complete explanation of the role this extra chromosome plays, see the accompanying section, "Chromosomes and Klinefelter syndrome." In the early 1970s, researchers around the world sought to identify males having the extra chromosome by screening large numbers of newborn babies. One of the largest of these studies, sponsored by the National Institute of Child Health and Human Development (NICHD), checked the chromosomes of more than 40,000 infants. Based on these studies, the XXY chromosome arrangement appears to be one of the most common genetic abnormalities known, occurring as frequently as 1 in 500 to 1 in 1,000 male births. Although the syndrome's cause, an extra sex chromosome, is widespread, the syndrome itself-the set of symptoms and characteristics that may result from having the extra chromosome-is uncommon. Many men live out their lives without ever even suspecting that they have an additional chromosome.

Definition: Klinefelter syndrome is a chromosome abnormality that affects only men and causes hypogonadism
Alternative Names: 47 X-X-Y syndrome
Causes, incidence, and risk factors: A person's sex is determined by the X and Y chromosomes. Normally, men have an X and a Y and women have two X's. In Klinefelter syndrome, a male has two X's and a Y. The condition is common and affects 1 in 500 men. The infant appears normal at birth, but the defect usually becomes apparent in puberty when secondary sexual characteristics fail to develop (or develop late). At this time, testicular changes occur that eventually result in infertility in the majority of those affected. Some mild cases may go undetected, with no abnormalities present except infertility. One risk factor for this condition is the mother giving birth at an older age. In most cases, the problem develops during fetal development and is not inherited from one or both parents.

Klinefelter Syndrome
Klinefelter Syndrome occurs in 1 in 500 to 1 in 1000 live births. People with this disorder develop as males with subtle characteristics that become apparent during puberty. They are often tall and usually do not develop secondary sex characteristics such as facial hair, or underarm and pubic hair. Men and boys with Klinefelter Syndrome have a Y chromosome and 2 X chromosomes. This is an example of trisomy Where does the extra chromosome come from? In about half of Kleinfelter cases, the extra X chromosome is from the mother's egg, while in the other half of cases the extra chromosome is from the father's sperm.

LIVING WITH KLINEFELTER SYNDROME My name is Stefan, I was diagnosed with Klinefelter Syndrome (KS) in February 1996 at the age of 25. Being diagnosed with KS has been, for the most part, a big sigh of relief after a life of frustrations. Granted, it was difficult and had a period of adjustment especially after finding out at age 25. Klinefelter Syndrome, as you may or may not know, is hard to diagnose. It occurs in roughly 1 in 500 males. First, I would like to tell you a bit about my life, mainly before the diagnosis. I was born in March of 1970. I was a happy baby, at least indicated by my parents and other family members. I was also told that I behaved well and did what I was told. However, it always seemed throughout my early childhood, teens and even somewhat now that I was very shy, reserved and had trouble making friends among my peers. My temper always got the best of me and I would fly into rages for no apparent reason. One minute I’d be very calm, the next I would be picking fights and throwing things. My parents knew when I was very young, that there was something about me that wasn’t right. But they could never figure it out until the diagnosis of Klinefelter Syndrome in February 1996.

Klinefelter Syndrome is probably the most common chromosomal variation found in humans. In random surveys, it is found to appear about once in every 500 to one in every 1,000 live born males. Since the largest percentage of these men would have never been diagnosed otherwise, it shows that in many cases affected individuals lead healthy, normal lives with no particular medical or social questions. Klinefelter Syndrome is diagnosed through a karyotype; a chromosome analysis usually done on a blood sample. Klinefelter Syndrome is caused by a chromosome variation involving the sex chromosomes. The person with Klinefelter Syndrome is a male who, because of this chromosome variation, has a hormone imbalance. While Dr. Harry Klinefelter accurately described this condition in 1942 , it was not until 1956 that other researchers reported that many boys with this description had 47 chromosomes in each cell of their bodies instead of the usual number of 46. This extra sex (X) chromosome causes the distinctive make-up of these boys. All men have one X chromosome and one Y chromosome, but sometimes a variation will result in a male with an extra X. This is Klinefelter Syndrome and is often written as 47,XXY. There are other, less common variations such as 48,XXYY; 48,XXXY; 49,XXXXY ; and XY/XXY mosaic. All of these are considered Klinefelter Syndrome variants.

A Parent's View
Boys are made of special things and sometimes some Xtra special things! Allow me to begin by telling you that many doctors and researchers do not refer to Klinefelter Syndrome when referring to a child because a child can not and does not have the symptoms of what Harry Klinefelter researched and not all babies and boys will go on to develop the symptoms associated with Klinefelters Syndrome and until they do, they do not have a syndrome. There is the blessing of the early diagnosis. SO much can be changed but just having that advantage. Use it to yours!! So you have just been told that your baby has an extraX.......There is so much more to know than just that! Please take the time to educate yourself. The unknown is a scary place to be and usually the FACTS are not as bad as they first may seem. Please read on!
I was in your shoes just a short time ago when I was told that my two month old baby had this disorder. I was terrified....but I also love my child with all of my heart so I took the time to learn. I will share with you what I felt, heard, read and learned and hopefully provide you with something I wished I had had available to me the day I was told.

In the early 1970s, researchers around the world sought to identify males having the extra chromosome by screening large numbers of newborn babies. One of the largest of these studies, sponsored by the National Institute of Child Health and Human Development (NICHD), checked the chromosomes of more than 40,000 infants.

A Parent's View
Boys are made of special things and sometimes some Xtra special things! Allow me to begin by telling you that many doctors and researchers do not refer to Klinefelter Syndrome when referring to a child because a child can not and does not have the symptoms of what Harry Klinefelter researched and not all babies and boys will go on to develop the symptoms associated with Klinefelters Syndrome and until they do, they do not have a syndrome. There is the blessing of the early diagnosis. SO much can be changed but just having that advantage. Use it to yours!! So you have just been told that your baby has an extraX.......There is so much more to know than just that! Please take the time to educate yourself. The unknown is a scary place to be and usually the FACTS are not as bad as they first may seem. Please read on!
I was in your shoes just a short time ago when I was told that my two month old baby had this disorder. I was terrified....but I also love my child with all of my heart so I took the time to learn. I will share with you what I felt, heard, read and learned and hopefully provide you with something I wished I had had available to me the day I was told.
