  • Cancer is the leading cause of death by disease in children between ages one and 14. An estimated 9,100 new cases are expected to occur this year in this age group, with an estimated 1,400 deaths. A cutting-edge oral anticancer drug now being tested in children 18 and younger with solid tumors is designed to stop the uncontrolled growth of cancer. The "epidermal growth factor receptor tyrosine kinase inhibitor" works by blocking a signal that drives the growth of many solid tumors.
    Asthma is the leading serious chronic disease among children. At least 7.7 million children under 18 have been diagnosed with asthma in the U.S. Asthma generated 658,000 emergency room visits in 1999 and is the leading cause of hospitalization among children under 15. It causes 10 million lost school days each year. A new medicine for asthma is an injected monoclonal antibody that blocks the chemical chain of events that triggers asthma. In clinical trials, the medicine sharply reduced asthma attacks in both children and adults.
    Fabrys disease is a rare genetic disorder affecting people who lack an enzyme that clears a fatty substance from the blood vessels of the heart, kidneys, skin and other organs. The buildup of the lipid often leads to reduced blood flow and organ failure, particularly in the kidneys, heart and brain. Most patients die in their forties. But a new medicine, a genetically engineered version of the missing enzyme, successfully cleared the fatty deposits that clogged the kidneys, heart and skin of the majority of the patients in a clinical trial.

It is characterized by deposition of a lipid, ceramide trihexose, in various tissues, sometimes leading to death from renal failure and hypertension in midlife. In addition to skin lesions, renal abnormalities, cardiomegaly and cerebral haemorrhage, patients may exhibit musculoskeletal manifestations; these may include osteonecrosis at many sites and periarticular swellings in elbows, knees and small joints of the fingers.

Other characters Fabry's disease, (Johannes Fabry, 18601930, German dermatologist), lysosomal disease of the group of sphingolipidoses clinically characterized by attacks of pain in the extremities and abdomen, triggered by exercise, fatigue and emotional stress. Ocular symptoms with corneal opacities and cataract subsequently appear, as does telangiectasis affecting mainly the inferior limbs. Cardiovascular symptoms appear with increasing age; transient ischaemic attacks, ischaemic infarctions, aneurysm formation, hemiplegia, aphasia, seizures and mental deterioration are part of the neurological picture. The diagnosis is made by demonstrating deficiency of alpha-galactosidase activity in plasma, leukocytes, urine and cultured skin fibroblasts. The basic defect is accumulation of glycosphingolipids in lysosomes. MR shows nonspecific small lacunar infarcts in both the white and grey matter, atrophy and occasional haemorrhages.

What is the effectiveness of a -galactosidase A in the treatment of Fabry's Disease? Fabry's disease is a rare genetic disorder of lipid metabolism characterised by deficiency of the enzyme alpha-galactosidase A which leads to the accumulation of fatty deposits within various organs in the body. Symptoms of the disease include abdominal pain and visual impairment. Later in the course of the disease there maybe serious complications due to kidney failure, heart irregularities and progressive neurological abnormalities. Fabry's Disease Subjective
A 67-year-old white female presented for a routine eye examination with typical visual complaints of occasional blur at distance and near. Her current eyeglasses are four years old. She has experienced mild photophobia in bright light and while driving at night. The patient is taking ranitidine hydrochloride (Zantac) and loratadine (Claritin D) and is allergic to ibuprofen (Motrin). There is no known personal or family history of ocular disease, surgery or injury.
Objective:
  • Visual acuity: (20/25) OU
  • Pupils: equal, round and reative OU
  • Lids: uninvolved OU
  • Conjunctiva: slight injection OU
  • Cornea: both corneas demonstrated a pronounced browish-tan verticillate pattern within the sub-epithelial tissues ( Fig.1 ). There was no disruption of precorneal tear film as demonstrated with flurescein dye
