Link between Epstein-Barr virus and multiple sclerosis Reported by Susan Aldridge, PhD, medical journalist Infection with the Epstein-Barr virus is linked with the emergence of multiple sclerosis many years later.
Previous research has suggested an association between Epstein-Barr virus (EBV) and multiple sclerosis (MS), a disabling disease of the nervous system. Researchers at the US Army Physical Disability Agency, Washington, sought to determine the time lag between infection and onset of MS symptoms. They analysed data from more than three million military personnel, all of whom had given blood samples, focussing on 83 people granted disability because of MS. Each case was matched with two healthy controls of the same age, sex and date of blood sample collection. Analysis of blood samples showed higher levels of EBV antibody in the MS cases. The presence of high antibody levels is a marker of infection. The study showed that infection occurred five years or more before the onset of MS. Source

HealthScoutNews Reporter TUESDAY, March 25 (HealthScoutNews) A new study strengthens the case for a link between infection with the Epstein-Barr virus and multiple sclerosis, researchers say. The case is not yet proven, says Dr. Alberto Ascherio, an associate professor of epidemiology at the Harvard School of Public Health who has worked to establish that link over the past few years. Ascherio is the lead author of a paper on the study in the March 26 issue of the Journal of the American Medical Association "We cannot say definitely, but this paper combined with the previous evidence does provide support for a causal relationship," Ascherio says. Epstein-Barr virus, a member of the herpesvirus family, is ubiquitous. Experts say up to 95 percent of Americans are infected with it by age 40. It has been linked to a number of other diseases, most prominently mononucleosis, and also several cancers, such as lymphomas. Ascherio speculates that the multiple sclerosis (MS)-virus link is due to an underlying genetic predisposition. In some individuals, he says, the virus somehow triggers an attack by the immune system on myelin, the protective covering of nerve fibers, causing problems ranging from numbness and tingling to paralysis.

Eng-Lai Tan Choon-Kook Sam Introduction Epstein-Barr virus (EBV) is associated with various types of malignancy, in particular, nasopharyngeal carcinoma (NPC). The viral oncogene, latent-membrane protein 1 (LMP-1), is known to transform B-lymphocytes and rodent fibroblast in vitro. It is thought to protect the infected cells from apoptosis by up-regulating the bcl2 and A20 genes. LMP-1 gene consists of three exons separated by short introns (Fig. 1). One of our investigation efforts attempted to study the possibility of using the LMP-1 protein as an antigenic marker for early progression of NPC.

All tumor-associated viruses have evolved strategies for survival and spread as part of their physiologic infection, which affects cell cycle and apoptosis control. By inducing transient proliferation in their target organ/cells they prepare for lytic replication by engaging more cells, as an amplification mechanism. Virus infections per se induce apoptosis, which may be regarded as a defence mechanism by the host/host cell. Progression from cells at risk to overt tumor only takes place if late replicative events are truncated or blocked (e.g. by integration, accidental infection of wrong cell type), if the infection cannot be handled by the immune system and/or if the infection raises an inappropriate host reaction.
Epstein-Barr virus (EBV) infection in man, illustrates these points, due to extenisve though incomplete knowledge on several of these issues. EBV is the most common virus in the human population (>90% of adults carry the virus), it normally gives rise to an inconspicuous or benign infection, paradoxically, however, under rare circumstances it induces tumors and in vitro it is very efficient in transforming its natural host cell, the B-lymphocyte.

ABSTRACT Although 5–10% of gastric carcinoma (GC) cases worldwide are associated with EBV, a human herpesvirus, it is still not clear what the precise contribution of the virus is to the pathogenesis of EBV-positive GC. Here we report that EBV infection induces expression of insulin-like growth factor 1 (IGF-I) in the GC-derived EBV-negative cell line NU-GC-3 and that the secreted IGF-I acts as an autocrine growth factor.
