Human Ebola vaccine trial begins

The leader of the team that designed an Ebola vaccine and tested it in macaque monkeys expects to see no adverse effects in the human trial The subjects will be monitored for 1 year, but Nabel anticipates no adverse effects, because other DNA vaccines have already proven safe. Plus, the vaccine contains no infectious materials from Ebola virus. Instead, he told The Scientist The current trial is just the first test of a vaccine that Nabel's group described 3 years ago in a report to Nature , detailing the first successful test of an Ebola vaccine in a nonhuman primate. That trial employed a prime/boost strategy. Once a month for 3 months, macaque monkeys were injected with plasmid DNA encoding Ebola virus glycoprotein and nucleoprotein to prime the immune system. Three months later (5 months after the first DNA injection), the monkeys received an injection of crippled adenovirus containing the glycoprotein gene to boost the immune response. The team then challenged the vaccinated animals 3 months later with a lethal dose of Ebola virus (at 8 months). Significantly, the vaccine induced both humoral and cellular immune responses in the animals. For instance, recognizing that the prime/boost strategy's multipart regimen could prove impractical, Nabel's team in August described an accelerated vaccine in another

Uganda issues Ebola alert
KAMPALA, Uganda Uganda has issued an Ebola virus alert after the deaths of 28 people from haemorrhaging near the border with the Democratic Republic of Congo. Sam Okware, Uganda's Ebola task force chairman, told Reuters on Monday: "We have been hearing of rumours of an outbreak of haemorrhagic fever in Congo and Gabon so we have put the districts on alert just in case it is Ebola." Doctors fear an outbreak of fever in the Congolese village of Misangandu, roughly 1,000 km (620 miles) southwest of Uganda, could be the dreaded Ebola virus. The World Health Organisation said at least 10 other people, including a nurse, had died of a mystery illness in the west African country of Gabon and attributed one of the deaths to the deadly virus. The WHO said in a weekly bulletin in Geneva on Friday that it had received reports of cases of suspected viral haemorrhagic fever which includes Ebola in Ogooue Iveindo province in the northeastern part of Gabon.

Ebola Virus in Zaire
Beginning with the January 1995 outbreak of Ebola virus in Kikwit, Zaire, the CHAART staff have been working with investigators from the Special Pathogens Branch at the Centers for Disease Control and Prevention ( CDC ) in Atlanta to generate images of the area where the first Ebola cases were found. The investigation in Kikwit was initially hampered to a great extent by the lack of accurate and up-to-date maps. The CDC team expressed a need to (1) accurately identify the location of exposure and residence of human patients, (2) geographically reference the collection of vertebrate and invertebrate materials from the ecological investigation, and (3) correlate these collections with underlying landscape and faunal characteristics. The long-term goals of the collaboration are to use remotely sensed environmental information to supplement observations of the basic ecology of the field sites, and to utilize a geographic information system to relate these data with other epidemiological information to investigate the temporal and spatial elements of the outbreak. As part of this collaboration, CHAART scientists have provided the CDC team with several image products, generated from archived SPOT multispectral data and current Landsat Thematic Mapper data . Experts: Slower rate of Ebola infections
Story Tools BRAZZAVILLE, Republic of Congo (AP) An Ebola outbreak in Republic of Congo has killed 79 people but appears to be slowing down, health investigators said Wednesday. The rate of new infections has started tapering off in the quarantined region of Cuvette West, where the deaths have occurred, World Health Organization spokesman Iain Simpson said in Geneva. Last week, health officials registered only eight new cases compared to as many as 25 in previous weeks. Nearly 100 people are still battling the disease. "I'm not suggesting that we won't see more cases, because we probably will," Simpson said. "But we're hopeful that the number of new cases will still drop." The viral disease is one of the world's deadliest, causing rapid death through massive blood loss in up to 90 percent of those infected. Ebola spreads through bodily fluids. Primates, hunted by many central Africans for food, can carry the infection. Villagers believing the disease to be a curse initially fled from emergency medical workers earlier this month.

L'Ebola è un virus in grado di provocare gravi febbri emorragiche. Più precisamente si tratta di un filovirus , dalla caratteristica forma allungata, che deve il suo nome al fiume della Repubblica Democratica del Congo dove fu isolato per la prima volta nel 1976. Dei quattro ceppi del virus isolati fino a ora, tre sono letali per gli esseri umani. Probabilmente il contagio alla nostra specie è avvenuto dalle scimmie e da qualche altro mammifero della foresta africana, ma l'origine e la modalità di trasmissione rimangono un mistero. A oggi si sono registrate quattro epidemie di Ebola: nello Zaire, nel Sudan, nel Gabon e nella Costa d'Avorio. La mortalità ha raggiunto l'88% dei casi rilevati. Il contagio avviene con il contatto diretto con il sangue (spesso tramite siringhe infette), il seme, le secrezioni e il vomito di chi è già infetto, di norma circa 4-16 giorni dopo l'esposizione all'agente virale. I primi sintomi sono febbre, vomito ed eruzioni cutanee: il virus attacca preferibilmente i reni, il fegato e la milza, sedi di emorragie interne. Le perdite emorragiche dopo pochi giorni sono generalizzate a tutti i vasi arteriosi e il sangue fuoriesce da occhi, naso e orecchie. La disgregazione dei tessuti interna provoca l'ultimo terribile sintomo, la fuoriuscita di liquido nero.

WEB EXCLUSIVE Bill Delaney: Ebola scare in Canada HAMILTON, Ontario Health officials are awaiting test results to determine if a woman who traveled to Canada from Africa is ill with the Ebola virus. Doctors at Henderson Hospital have run a complete battery of tests on the woman, including tests for other hemorrhagic fevers, but said in a news conference they may never find out what disease the woman is suffering from. "We many never know" what she has, said Dr. Douglas MacPherson of Health Canada, the national health department. "But in terms of the diagnostic testing, we will have a yes/no type response certainly out of Winnipeg, within the two or three day frame that we were talking about," he added. If the woman turns out to have the Ebola virus, it would be the first known case of the often-fatal disease in North America.

WASHINGTON (Reuters) The deadly Ebola virus, which emerged mysteriously from African forests, probably attacks people who butcher and eat infected animals, researchers said on Thursday. The virus, which most recently killed 29 people in the Congo Republic, seems to break out when people slaughter chimpanzees, gorillas and small antelopes called duikers, the scientists said. "Humans and duikers scavenging for meat probably became infected by contact with dead apes," they wrote in their report, published in Friday's issue of the journal Science. "Almost all human Ebola outbreaks in Gabon and the Republic of Congo have been linked to the handling of dead animals by villagers or hunters, and increased animal mortality always preceded the first human cases," added the international team of researchers, led by Eric Leroy of the Development Research Institute in Gabon. They said health workers may be able to get a warning of Ebola outbreaks when large numbers of dead animals start appearing in forests.

Filoviruses are string-shaped, with a little hook or loop at one end. Another filovirus is the Marbug virus, which gives similar symptoms to Ebola, but the chance of surving an infection of Marbug virus is higher.
