What is Batten disease?
Batten disease is a fatal, inherited disorder of the nervous system that begins in childhood. Early symptoms of this disorder usually appear between the ages of 5 and 10, when parents or physicians may notice a previously normal child has begun to develop vision problems or seizures. In some cases the early signs are subtle, taking the form of personality and behavior changes, slow learning, clumsiness, or stumbling. Over time, affected children suffer mental impairment, worsening seizures, and progressive loss of sight and motor skills. Eventually, children with Batten disease become blind, bedridden, and demented. Batten disease is often fatal by the late teens or twenties. Batten disease is named after the British pediatrician who first described it in 1903. Also known as Spielmeyer-Vogt-Sjogren-Batten disease, it is the most common form of a group of disorders called neuronal ceroid lipofuscinoses (or NCLs). Although Batten disease is usually regarded as the juvenile form of NCL, some physicians use the term Batten disease to describe all forms of NCL.

The condition is named after the British paediatrician who first described it in 1903. Batten disease is relatively rare occurring in about one in 30,000 births. The illness leads to a progressive deterioration of the brain and nervous system. Although our understanding of Batten disease is improving all the time, there is at present no cure or treatment that has any significant impact on the inexorable decline in bodily functions and inevitable early death. A number of different forms of Batten disease have been identified. They are classified by age but are all genetically different.

What is Batten disease?
Batten disease is a group of related neurometabolic diseases affecting children and young adults. The disease occurs in about one in 30,000 births and leads to a progressive deterioration of the brain and nervous system. At present, there is no cure or treatment that has any significant impact on the unstoppable decline in bodily functions.
Symptoms may vary, but include sizures, dementia, blindness, loss of motor abilities and muscle coordiantion, and mental deterioration. Many affected die in childhood. The BDFA strives to:

General Discussion Batten Disease is the juvenile form of a group of progressive neurological diseases known as neuronal ceroid lipofuscinoses (NCL). It is characterized by accumulation of a fatty substance (lipopigment) in the brain as well as in tissue that does not contain nerve cells. This lipopigment storage disorder is inherited, and is marked by rapidly progressive vision failure (optic atrophy), and neurological disturbances, which may begin before eight years of age. Occurring mostly in families of Northern European Scandinavian ancestry, the disorder affects the brain and may cause both deterioration of intellect and neurological functions.

Batten disease
JNCL is caused by mutation called CLN3, which is located on chromosome 16p12.1. The CLN3 gene encodes a predicted transmembrane 438 amino acid protein called battenin the function of which remains unknown. While 31 different CLN3 mutations have been found, in most cases of JNCL disease, chromosomes carry the inherited defect a 1.02 kb deletion(exon 7 and 8) of genomic DNA, resulting in a truncation of CLN3 such that the resultant protein contains the original 153 N-terminal residues followed by 28 novel amino acids .
In affected individuals, mutations of CLN3 result in pathologic intracellular inclusions in neurons and other cells including liver, lymphosytes and fibroblasts. Lysosomal accumulation of autofluorescent lipids and high concentrations of a mitochondrial protein, ATP synthase subunit C, have been reported. Recent immunofluorescence studies have shown that the predominant intracellular destination of nascent CLN3 is the lysosome .
Cln3-knockout mice homozygous for a targeted deletion of exons 1?6 in the Cln3 gene have been previously reported to show characteristic accumulation of autofluorescent lipopigments containing mitochondrial ATP synthase subunit c in neural tissue, and selective loss of GABAergic neurons.
