General Discussion
Leigh's Disease is a rare inherited neurometabolic disorder. It is characterized by the degeneration of the central nervous system (i.e., brain, spinal cord, and optic nerve). The symptoms of Leigh's Disease usually begin between the ages of three months and two years. Symptoms are associated with progressive neurological deterioration and may include loss of previously acquired motor skills, loss of appetite, vomiting, irritability, and/or seizure activity. As Leigh's Disease progresses, symptoms may also include generalized weakness, lack of muscle tone, and episodes of lactic acidosis, which may lead to impairment of respiratory and kidney function. In most cases, Leigh's Disease is inherited as an autosomal recessive genetic trait. However, autosomal dominant, X-linked recessive, and mitochondrial inheritance have also been noted. There appear to be several different types of genetically determined enzyme defects that can cause Leigh's Disease.

What is Alpers' Disease?
Alpers' disease is a rare, progressive neurodegenerative disease of the brain that occurs in infants and children. It is an autosomal recessive disorder that is sometimes seen in siblings. First signs of the disease, which include intractable seizures and failure to meet meaningful developmental milestones, usually occur in infancy. Primary symptoms of the disease are developmental delay, progressive mental retardation, hypotonia (low muscle tone), spasticity (stiffness of the limbs), and dementia. Seizures may include epilepsia partialis continua, a type of seizure that consists of repeated myoclonic (muscle) jerks. Optic atrophy may also occur, often leading to blindness. And, although physical signs of chronic liver dysfunction may not be present, many patients suffer liver impairment leading to liver failure. While some researchers believe that Alpers' disease is caused by an underlying metabolic defect, no consistent defect has been identified. Is there any treatment?
