Op eigen gelegenheid had hij op de middelbare school al H.A. Lorentz' Beginselen der Natuurkunde (1888/1890) bestudeerd, en in Leiden verdiepte hij zich zelfstandig in de geschriften van L.E. Boltzmann, J.C. Maxwell en J.W. Gibbs. Tot echt fysisch inzicht kwam hij echter pas na lezing van het lange artikel dat Paul Ehrenfest en zijn vrouw Tatiana in 1911 over de statistische mechanica hadden gepubliceerd in de . Vanaf dat moment concentreerde Uhlenbeck zich op deze tak van de natuurkunde. Mededeelingen van het Nederlandsch Historisch Instituut te Rome (4 (1924) 217-228), handelend over de Nederlander die in 1603 tot de oprichters van de Accademia dei Lincei behoorde. Toen Uhlenbeck in juni 1925 definitief naar Nederland terugkeerde, had hij besloten geschiedenis te gaan studeren. De Leidse hoogleraar oud-Germaans C.C. Uhlenbeck, een neef van zijn vader, wees hem erop dat hij daarvoor Grieks en Latijn zou moeten leren. Bovendien raadde hij hem aan eerst in de natuurkunde te promoveren, nu hij toch al zover was. Uhlenbeck nam deze adviezen ter harte. Hij ging lessen in de klassieke talen volgen en begon terzelfder tijd bij Ehrenfest - wiens assistent hij werd - aan een dissertatieonderzoek.

Doctorado en 1927 por la Universidad de Leyden, emigró a los Estados Uni­dos ocupando un puesto de profesor de la Universidad de Michigan. En el transcurso de la Segunda Guerra Mundial participó en el desarrollo del radar. Desde 1961 formó parte del Ins­tituto Rockefeller de Nueva York. Junto con Samuel Abraham Goudsmit interpretó el cuarto número cuántico (postulado por Wolfgang Pauli para la completa definición de los átomos) como un giro del electrón, demos­trando que éste podía adoptar los va­lores + 1/2 o - 1/2.
