In 1494 Pacioli wrote his fifth book, Summa de Arithmetica, Geometria, Proportioni et Proportionalita (Everything About Arithmetic, Geometry and Proportion), a digest and guide to existing mathematical knowledge. Its 36 chapters on double-entry bookkeeping constituted one of five topics covered and were included, in part, to "give the trader without delay information as to his assets and liabilities." The Summa's publication on November 10, 1494 made Pacioli instantly famous, becoming the most widely read mathematical work in Italy and one of the first books published on the Gutenberg press (named for the German printer, the inventor of movable type). After the Summa's publication, Pacioli was invited to Milan to teach mathematics where one of his pupils was Leonardo da Vinci, to whom he taught perspective and proportionality. This knowledge allowed da Vinci to create one of his greatest masterpieces, and the most famous painting of the fifteenth century, a mural on the north wall of the Santa Maria de Gracia Dominican cloister known as "The Last Supper." In 1514, Pope Leo III called Pacioli to the papacy in Rome to be a teacher. Scholars believe Pacioli never made it to Rome, dying on June 19, 1517 in the monastery in Sansepulcro. In the first century after its publication, the Summa was translated into five languages and numerous details of bookkeeping set forth by Pacioli were followed in texts and the profession for at least the next four centuries.

Luca Pacioli gilt als einer der bedeutendsten Mathematiker des 15. Jahrhunderts - allerdings mehr auf Grund seiner kompilatorischen Arbeiten und seiner Lehrtätigkeit als wegen seiner eigenen Beiträge zur Entwicklung der Mathematik. Pacioli erfüllte vor allem die Funktion mittelalterlicher Kopisten, die die überlieferten Kenntnisse von einer Generation an die nächste weitergaben. Obwohl sein Beitrag zur mathematischen Entwicklung marginal ist, war seine Tätigkeit als Lehrer und Vermittler von großer Bedeutung für die Mathematik. Die Summa Paciolis steht an der Nahtstelle zweier Epochen: mit ihrer Gliederung und ihrem enzyklopädischen Ziel, das gesamte Wissen über ein bestimmtes Gebiet abzudecken, steht sie in der mittelalterlichen Tradition der Scholastik, während auf der anderen Seite die Neigung des Autors zu spezialisiertem Fachwissen und die neue Bewertung der Rolle der Mathematik in Wissenschaft, Technik und Kunst die Summa als ein Werk der Renaissance erscheinen lassen. Die Schrift faßt und organisiert die mathematischen Kenntnisse des Abendlandes seit Fibonacci zusammen, womit er den Ausgangspunkt ihrer weiteren Entwicklung lieferte. A circle is drawn on this vertical with the diameter given by the height of the picture. This circle intersects the pointing stick exactly where it meets the upper edge of the chalkboard. 3. Two regular pentagrams are inscribed in the circle, one "upright" and one "upside down". The point where they intersect on the horizontal diameter of the circle is the same point at which the pointing stick cuts it. Thus the stick is exactly positioned to define the two pentagrams. The sides of the pentagrams pass through Pacioli's right eye, the two outer knots on his belt, the gap between his 2nd and 3rd fingers on his left hand, the end of the "ink-cylinder" and the clothing of P and his mysterious friend.
