Evolution (BISC 400) Course page Current Research Program My research focuses on the importance of ecological selection pressures for the evolution of plant traits. Of primary interest is mating system evolution; in addition, I study aspects of plant-insect interactions, including both pollination and herbivory. My approach has been to test current models of trait evolution within the context of the natural history of an organism. I choose traits that have a genetic basis that is known or can be determined (using classic quantitative genetic techniques), and so can respond to selection; combine controlled experiments with studies of natural populations; and where possible use genetic markers to refine my estimates of realized reproductive success (e.g. paternity analysis and outcrossing rate estimation). To date, I have used this combined methodology to study the sex-specific fitness consequences of variation in flower size and number in horsenettle, Solanum carolinense , and to study the maintenance of an herbivore resistance polymorphism in western jimsonweed

Extractions: References An initial genetic dissection of ovule development In a second step we wanted to approach organogenesis at the functional level. One way to do this is to dissect genetically the process and to study systematically a large number of mutants. Hence, in order to identify the key regulatory factors involved in the above outlined developmental processes we have performed a large-scale single-line EMS mutagenesis to screen for sterile plants with defects in ovule development ( Schneitz et al, 1997 ). A total of 458 independent mutants were isolated. A few of these mutants have been independently isolated in other laboratories as well (Baker et al, 1997; Elliott et al, 1996; Gaiser et al, 1995; Klucher et al, 1996; Modrusan et al, 1994; Robinson-Beers et al, 1992). An initial analysis identified several groups of genes that control different aspects of ovule ontogenesis. Some appear to be important for early processes. For example, the

Extractions: Our research focuses on the genetic regulation of vascular tissue development in plants. Specific questions are (1) what is the molecular mechanism behind vascular strand formation? (2) which genes regulate fiber differentiation? (3) which genes regulate the rate of wood formation and the cellular composition of wood? We focus our research on two model plant species, Arabidopsis and poplar, since considerable genetic resources are available for both species. In Arabidopsis we are using both traditional mutagenesis and reverse genetics to approach gene function, whereas reverse genetics is the preferred choice in poplar.
