I have developed and applied skills in microbiology and infection control , supervised as many as 26 employees in large (500 bed hospitals) reference laboratories. My experience includes all areas of microbiology to include virology (both serology and viral isolation), parasitology mycology tuberculosis , and clinical immunology . I taught in active training programs in these centers for many specialties to include infectious disease pathology , and medical technology . I have been an invited lecturer at University of Texas Health Science Center in San Antonio, and I am a contributing author to the textbook used in that program.
I have built complex relational databases in Access and Paradox for management of laboratory supply operations. I have designed a laboratory computer system featuring edit checks and conditional branching logic. I was a reviewer at the Center for Drug Evaluation and Research - Anti-Infectives at the Food and Drug Administration . I have written many computer applications that improved operations, and cut costs. I have used

26. Haemolytic Anaemia Testing
Tests to check for the presence of hemolytic anemia.
http://www.uq.edu.au/vdu/HDUHaemolyticAnaemiaTests.htm
Haemolytic Anaemia Testing To check for the presence of any haemolytic anaemia the following test may be performed: Haematology Profile (FBC/FBE)
  • Should demonstrate an anaemia
Blood Film
  • Schistocytes
    • Products of RBC breakdown
    • Suggests intravascular haemolysis (IVH)
  • Microspherocytes
    • Intra/Extravascular haemolysis (IVH/EVH)
    • A hallmark of haemolytic anaemia (HA)
  • Polychromasia (IVH/EVH)
  • Nucleated Red Blood Cells (IVH/EVH)
  • Erythroid hyperplasia is more pronounced than in HA than in non-HA (Dec. M:E)
  • Noticeable that the lifespan of RBCs from a patient with an intrinsic RBC defect, when infused into a normal patient, are decreased. Normal RBCs infused into a patient with an intrinsic RBC defect do not haemolyse.
Osmotic Fragility Test (OFT)
  • A measure of the RBCs resistance to haemolysis by osmotic stress, depending primarily on the volume of the cell, the surface area and the cell's membrane function
  • RBCs are incubated in varying concentrations of a hypotonic solution of sodium chloride (NaCl)
  • As the concentration of NaCl decreases, cells take on water to produce osmotic equilibrium

27. Molecular Virology Lab At Leiden University
The group performs basic research on viruses in order to facilitate the development of efficient genetransfer vectors for use in gene therapy. Current research concerns the human adenoviruses and the systems required for producing recombinant-adenovirus vectors.

E-mail Webmaster Virus Biology lab. Last modified: 03-03-2004
The Virus Biology lab. is an academic-research group embedded in the Department of Molecular Cell Biology at Leiden University Medical Center , Leiden, The Netherlands. The group performs basic research on viruses in order to develop efficient virus-derived gene-transfer vectors for use in e.g. gene therapy. In their evolution, viruses have become very efficient in transferring genetic information into cells. We study the various mechanisms exploited by viruses to do so. Our current research centers around adenoviruses, lentiviruses, and circoviruses.
Our Ambition:
To become a leading provider of new virus-derived gene-transfer tools for preclinical and clinical use in biomedical research and experimental gene therapy.
The Labs Profile:
The goal of our research is to develop and characterize efficient virus-derived gene-transfer vectors for biomedical research in general and experimental gene therapy in particular. Therefore we study the mechanisms by which viruses transfer their genes into cells and how they evade the immune system. Currently we focus on adenoviruses and lentiviruses as vectors for gene transfer. Click here for more information.

28. Megaloblastic Anaemia
Haematological profile, diagnosis and treatment of megaloblastic anemia. Vitamin B12 deficiency and frolic acid deficiency.
http://www.uq.edu.au/vdu/HDUAnaemiaMegaloblastic.htm
Megaloblastic Anaemia Diagnosis
  • Many cases are seen that have a decreased serum B12 with a normal haematological profile and possibly normal clinical findings
  • Test for methylmalonic acid in the serum as an indicator of vitB12 deficiency
  • Test for homocysteine to check folate levels
Haematological Profile
  • Hb/RCC/Hct - Dec (due to Dec. DNA production)
  • MCV/MCH - Inc. (As precursors are megaloblastic)
  • MCHC - Normal
  • RDW - Inc. (Anisocytosis due to microcytes, normocytes and oval macrocytes)
  • Plt./WCC - Dec
  • Pancytopaenia with a normochromic, macrocytic anaemia
  • Intramedullary haemolysis due to ineffective erythropoiesis (reason for hypercellular BM)
  • Poikilocytosis - ++ (tear drops and schistocytes)
  • Anisocytosis - +++ (oval macrocytes)
    • A "Shift-to-the-Left" indicates an Inc. in band form WBCs
  • Hypercellular bone marrow (M:E at 1:1 or reversed)
  • Erythron
    • Large cells with fine chromatin (Megaloblasts)
    • Nuclear(N)/Cytoplasmic(C) dissociation with N lagging C - normal cytoplasmic maturation
    • Multiple Howell-Jolly bodies (diagnostic if observed)
  • Leucon
    • Giant myelocytes and metamyelocytes
  • Thrombon
    • Often not obvious
  • The peripheral blood (PB) and bone marrow (BM) findings do not distinguish between a vitamin B12/cobalamin deficiency and a folic acid (folate) deficiency
