    Petr Karlovsky lab: Molecular Phytopathology
    We study plant pathogenic fungi and their interactions with host plants and with competing and antagonistic microbes using tools of molecular genetics and biochemistry. The primary focus of our research are secondary metabolites. Some fungal metabolites are involved in pathogenicity as virulence factors or suppressors of plant defense reaction, others play a role in interactions with competing or mycoparasitic fungi . Because of their function in pathogenicity (virulence factors) and toxic effects they exert on humans and animals (mycotoxins), secondary metabolites of phytopathogenic fungi affect both yield and quality in plant production. Molds and fungi are found in virtually every environment and can be detected, both indoors and outdoors, year round. The number of species of existing molds and fungi is estimated from tens of thousands to three hundred thousand or more. Molds and fungi produce and release millions of spores small enough to be air-, water-, or insect-borne. They can also produce toxic agents known as mycotoxins. Spores and mycotoxins can have negative effects on human health including allergic reactions, asthma and other respiratory problems.
    This page is maintained as a product of the alliance between OSHA and the American Biological Safety Association (ABSA). The following questions link to resources that provide safety and health information relative to molds and fungi.

Included are the conspicuous mushrooms , but also many microscopic forms such as molds and yeasts . Some 70,000 species have been described, and perhaps 1.5 million species actually exist, with the majority yet to be identified and described by mycologists (Hawksworth, 1991; Hawksworth et al., 1995). The fungi have undergone substantial changes in the arrangement of the various taxa in the last several decades, especially as techniques for comparing biochemical characteristics (such as ribosomal RNA and DNA) have become increasingly more sophisticated. The phylogeny presented here is after Bruns et al. (1991, 1993) for the Eumycota (true fungi) and recognizes four divisions: the Chytridiomycota (chytrids)

    General:
    1. To describe the dimorphic nature of the pathogenic fungi used in making a clinical diagnosis 2. To emphasize the eukaryotic nature of the fungi and its relationship to pathogenesis. 3. To establish familiarity with the scientific terminology peculiar to mycology. 4. To explore the nature of the pathogenesis of fungal infections. 5. To gain familiarity with the classification of medically-important fungi. 6. To develop an understanding of the nature and mode of action of anti-fungal agents.

    General Properties of Fungi
    Structural-functional relationships
    The fungi are more evolutionarily advanced forms of microorganisms, as compared to the prokaryotes (prions, viruses, bacteria). They are classified as eukaryotes , i.e., they have a diploid number of chromosomes and a nuclear membrane and have sterols in their plasma membrane. Genetic complexity allows morphologic complexity and thus these organisms have complex structural features that are used in speciation.

    FUNGI OF NOTE Armillaria ostoyae Ashbya gossypii is a source of vitamins, such as riboflavin. Aspergillus flavus produces a poison called aflatoxin on peanuts and other crops that can make people who eat these foods sick. Aspergillus niger makes enzymes used in laundry detergents and many other products, and for tanning leather. Beauveria bassiana is used as a pesticide to control nuisance insects. Candida albicans causes yeast infections. Fusarium is a group of fungi that cause diseases in a wide range of plants. Helminthosporium oryzae causes disease in rice and led to the Bengal famine of 1942.
