Introduction
Geology currently runs a JEOL JSM-840 Scanning Electron Microscope fitted with LINK EXL software giving us a wide range of analytical capability. Secondary and Backscattered Electron Imaging, X-Ray EDS microanalysis and X-Ray mapping are all possible. This Web page contains examples of the different types of data that can be obtained from the SEM, and represents the basis of the lecture "Principles of Scanning Electron Microscopy" given on Module 8343 - Geochemistry.
Secondary Electron Images - High Resolution Topographic Imaging
Secondary electrons are low energy electrons ejected from the sample as a result of inelastic collisions with beam electrons. They are produced near the surface of the sample, from a small area around the beam tip. The resolution of the image is therefore very good because the beam is only a few nanometres in diameter. The image below is a Secondary Electron Image of a microfossil. Because the electron beam carries a charge, the sample has been coated with a very thin coat of gold - this enables any charge to drain off to earth. The benefits of SEM over conventional microscopy include very high resolution and greater depth of field.
Backscattered Electron Images - Qualitative Compositional Information
Backscattered electrons are primary electrons emitted as a result of elastic collisions with specimen electrons. BSE emission intensity is a function of the specimen's atomic number; i.e., the higher the atomic number (e.g., Fe(26) vs. Mg(12)), the brighter the signal. BSE images are obtained in exactly the same way as Secondary Electron Images.

Photo: Eric Johnson As the author of this site, Miloš Kaláb welcomes you to view foods, particularly dairy products, as they appear at high magnifications under electron microscopes. Structure is a very important attribute of foods. Cheese, pork roast, bread, ice cream, and many other foods are characterized not only by their different compositions but more evidently by their microstructures. Interestingly, more than 30 years ago, the author was asked to investigate the feasibility of processing milk powder into a product resembling a wiener ("hot dog"). It became clear from several experiments that a food based on protein in globular form (milk) would require sophisticated technology to produce fibres which are characteristic of meat products, including wieners. Electron microscopy images obtained by a biologist captured the attention of Milos to such an extent that they have changed his career for the rest of his life. His biography lists some of his most important scientific papers.

General Information

The Electron Microscopy Laboratory is a state-of-the-art facility that provides powerful techniques for characterization of the microstructure and chemical composition of materials through electron imaging, electron diffraction, energy dispersive x-ray spectroscopy (EDS), and electron energy loss spectroscopy (EELS) from minimeter scale to nanometer or atomic level. This laboratory is a multi-user, interdisciplinary facility that is open to students, staff and faculty at Boston College, as well as to users and scientific collaborators from other institutions. Access to the instrument is by arrangement with Dr Jianyu Huang (Lab. Manager) or Professor Zhifeng Ren . For users who are interested in being trained to use the instrument themselves, we offer two continuous courses (Electron Microscopy I-Fundamental Theory, Fall Semester ; Electron Microscopy II-Experimental, Spring Semester ) at a graduate level.

UMaine's Electron Microscopy Laboratory (EML) offers the tools and technical expertise for research and training in microscopy with both light and electron microscopes. The lab operates three electron microscopes (one scanning and two transmission electron microscopes) and the ancillary equipment needed for preparation of specimens for EM and other microscopy. The EML's variety of light microscopes encompasses optics for bright-field, phase-contrast, differential-interference, and fluorescence imaging. Users of the EML can opt for full technical services in microscopy so that they are provided with whatever micrographs and analyses they want for given specimens, or they can work independently on the EML's microscopes, specimen-preparation equipment, and darkroom facilities following appropriate training by the EML staff.
