Electron Microscopy of Macromolecular Assemblies
M y laboratory studies the architecture of large biological complexes and the structural basis for their regulation. We are applying cryo-electron microscopy (cryo-EM) and image analysis to characterize the structure and assembly of microtubules and their interaction with cellular factors and antimitotic ligands. The atomic model of tubulin that we obtained by electron crystallography has allowed us to establish the structural basis of nucleotide exchange and polymerization-coupled hydrolysis. The high-resolution model of the microtubule, on the other hand, has allowed us to propose a mechanism for the hydrolysis-induced depolymerization of microtubules. We are now characterizing the conformational change in tubulin that triggers microtubule disassembly following GTP hydrolysis, as well as the interaction of tubulin with the cellular microtubule depolymerizer XKCM1. We are also pursuing the structures of the eukaryotic, general transcriptional machinery

    Welcome to my website
    Finally I started to set up my image gallery where I am presenting pictures of various lymphotropic herpesviruses using electron microscopy, immuno electron microscopy and immuno fluorescence microscopy. I am going to add more pictures every week or when I get time for scanning in the pictures. These pictures were taken on cell lines available in my laboratory and using monoclonal antibodies developed in my lab. The herpesvirus information page will be added soon. The Flash 4 program ( virtual microscope) for fluorescence and electronmicroscopic pictures will be added (finally it start to work), which will allow to view the pictures, presented in the image gallery, at various magnifications and to move around in the picture.     Manuscript Predating Watson and Crick Article Now Published in The Journal of Structural Biology
    The March 2004 issue of the Journal of Structural Biology features the publication of a manuscript describing a model of the structure of DNA built in 1951. Authored by King's College graduate student Bruce Fraser, the manuscript predates the seminal 1953 paper describing the double helix structure of DNA published by Watson and Crick, and was in fact, cited in that article as "in press". The Fraser manuscript, however, was never actually published. The work provided important information leading to the establishment of the double helix structure of DNA and is considered a significant and historic manuscript.

    What is Bugscope? Live Schedules The Bugscope project is an educational outreach program for K-12 classrooms. The project provides a resource to classrooms so that they may remotely operate a scanning electron microscope to image "bugs" at high magnification. The microscope is remotely controlled in real time from a classroom computer over the Internet using a web browser. Bugscope provides a state-of-the-art microscope resource for teachers that can be readily integrated into classroom activities. The classroom has ownership of the project - they design their own experiment and provide their own bugs to be imaged in the microscope. The Bugscope project is primarily oriented towards K-12 classrooms and there is no cost to participate in the project. If you would like to take part in the Bugscope project, our how to participate pages will guide you through the simple steps needed to apply, schedule a session and operate the microscope. Our other resources pages will provide helpful links related to electron microscopy and bugs.
