  • Factors Affecting the PCR
    Factors Affecting the PCR:
    Denaturing Temperature and time
The specific complementary association due to hydrogen bonding of single-stranded nucleic acids is referred to as "annealing": two complementary sequences will form hydrogen bonds between their complementary bases ( G to C, and A to T or U ) and form a stable double-stranded, anti-parallel "hybrid" molecule. One may make nucleic acid (NA) single-stranded for the purpose of annealing - if it is not single-stranded already, like most RNA viruses - by heating it to a point above the "melting temperature" of the double- or partially-double-stranded form, and then flash-cooling it: this ensures the "denatured" or separated strands do not re-anneal. Additionally, if the NA is heated in buffers of ionic strength lower than 150mM NaCl , the melting temperature is generally less than 100oC - which is why PCR works with denaturing temperatures of 91-97 o C A more detailed treatment of annealing / hybridisation is given in an accompanying page, together with explanations of calculations of complexity, conditions for annealing / hybridisation, etc.

http://hem.passagen.se/thebee/SCIENCE/cosmcell.htm
COSMOLOGICAL CELL BIOLOGY? In his lectures on the teaching of cell biology in the upper grades of the waldorf schools, Rudolf Steiner suggested that cell biology be described from a cosmological perspective. Cell structures
In his course on the relation of the different sciences to astronomy , when talking of cells, he mainly points to the cell as a cosmos and the inner relation between structures in the cell and structures in the solar system (solar systems) (first lecture).
Not so much was known in his days as today about the subtle and intricate inner structures of the cell. Much more can surely be developed in this field today. Cell biographies - cell cycles
Another perspective is however the temporal perspective on the life of the cell.
Here, I feel, it it much easier to come to an understanding and picture of the development of cells as a mirror of the development of cosmos. If you look at the cell cycle with its interphase and its division stages (mitosis and meiosis), it is difficult not to feel enjoyment at the beauty of its inner order. This is clear in a number of aspects for interphase and mitosis. I have not worked on with meiosis yet. THE PERSPECTIVE OF ISOLATED STAGES
This is the way one part of the life cycle of an animal cell; mitosis, rather normally is depicted in biology books. The picture is taken from "Cell and Molecular Biology" by Robertis and Robertis (1979, p 384). The accompanying text describes the stages depicted in its details.

Aims and Scope
Physiologia Plantarum has been published by the Scandinavian Society for Plant Physiology since 1948. The journal publishes original, hypothesis-driven research describing aspects of experimental plant biology covering molecular and cell biology, biochemistry and biophysics and ecophysiology. By maintaining this broad scope, Physiologia Plantarum aims to facilitate the exchange of knowledge between researchers working at different organizational levels of plant biology. Physiologia Plantarum has been using a fast, web-based manuscript handling and review platform since 2000, with a time to first decision for papers received in 2003 of around 6 weeks. To further speed up publication, in spring 2004 all papers accepted will be published electronically via

IMB is the leading national institution in the field of molecular biology, molecular genetics, cell biology, biochemistry and bioorganic chemistry. Its achievements in these and related areas are documented in several hundred scientific papers published in international journals. IMB participates in many international collaborative projects that proves the high quality of the research carried out in it. Many of the projects are financed by EC, ICGEB, NATO, UNESCO, and other international organizations.
Since 1990 IMB is a member of the Global Network for Molecular and Cell Biology (MCBN) at UNESCO.
