Background

The finding that tumor viruses were permanently associated with transformed cells raised the possibility that transformation might result from the expression of viral gene products. In the case of Rous sarcoma virus, the first key observation was made by Howard Temin in 1960. He showed that a mutant of Rous sarcoma virus could cause the production of morphologically distinct, fusiform or spindle-shaped cells, readily distinguishable from the rounded cells that resulted from infection by wild-type virus (Temin, 1960). Temin therefore concluded that the morphology of the transformed cell is controlled by a genetic property of the virus.

The isolation of temperature-sensitive mutants also made possible a detailed examination of the biochemical events that occur during transformation. Temperature-shift experiments showed that a number of membrane-associated events occurred early in the transformation process, and could occur in the absence of protein synthesis. In a classic experiment, Beug and Graf showed that cells infected with ts mutant virus could undergo morphological transformation even when enucleated, although later events were blocked by enucleation (Beug et al., 1978). The picture that emerged from these early studies was that the transformation was initiated at the plasma membrane - where the Src protein was found to reside - and that signaling pathways then conveyed signals to the nucleus.

    We would like to thank Djamel Tahi and Huw Christie for asking us to comment on Professor Luc Montagnier's answers given in his interview with Djamel Tahi. Before commenting we thought it would be useful to start with a brief review of the methods used to prove the existence of retroviruses, and Montagnier et al 1983 evidence for the existence of "HIV". Methods used to prove existence of retroviruses
    de novo in each tumour. There is no epidemiological evidence that cancer comes into the body from outside; everything we know supports the classical view that it is a local autochthonous disease. Experimental sarcomas produced by embryo extract and indol, arsenic or tar have been transmitted by filtrates. Epitheliomas are easily produced in mice by tar and in men by chronic irritation; and if we believe that all malignant tumours contain more or less of a carcinogenic agent akin to the Rous virus, it follows that we can with a considerable degree of certainty stimulate normal tissues to produce virus".(2) proviruses ... One more question is worth answering: What form would the mutant protein be likely to take in the tumour cell? On account of its rapid multiplication it might well show a higher degree of aggregation than its progenitor. It would then appear as an alien particle in the mutant cell. This is borne out by the electron microscope observations on two chicken tumour agents of provirus type by Claude, Porter and Pickels (1947)".(3)

    Soviet cosmonaut who piloted the Vostok 4 spacecraft, launched 12 Aug 1962. He and Andriyan G. Nikolayev, who was launched a day earlier in Vostok 3 , became the first two men to be in space simultaneously. He collected experimental data on the possibility of establishing a direct link between two space ships; coordination of astronauts' operations; and the effects of identical spaceflight conditions on the human organism. Problems with his life support system dropped the cabin temperature to 10 degrees Centigrade. He still managed to conduct experiments,and take colour motion pictures of the terminator between night and day. He subsequently commanded the flight of Soyouz 14 launched 3 Jul 1974 which docked with the Salyut 3 space station. Reinhard Selten

    German mathematician who shared the 1994 Nobel Prize for Economics with John F. Nash and John C. Harsanyi for their development of game theory, a branch of mathematics that examines rivalries among competitors with mixed interests. Selten achieved a decisive breakthrough in game theory: The introduction of the concepts of sub-game perfect and perfect equillibria reduced the set of Nash equillibria drastically by excluding threats that are not credible. Thus, more precise and sensible predictions can be made for many games, e.g. markets. Additionally, game theory has found applications in all of social sciences and even in biology. Pierre Dansereau
