Max Perutz e l'emoglobina
ono un chimico che lavora in un centro di ricerche di fisica su argomenti di biologia". Così amava definirsi Max Ferdinand Perutz, morto a Cambridge il 6 febbraio, a quasi 88 anni. Un chimico che ha avuto un'influenza profonda sulla medicina, svelando l'architettura e il meccanismo d'azione dell'emoglobina e ponendo così le basi della biologia molecolare. "Fu lui a insegnarmi che l'enigma della vita era racchiuso nella struttura delle proteine, e che la cristallografia a raggi X era il solo modo per venirne a capo" racconterà in seguito Perutz. All'epoca si pensava che anche i geni fossero costituiti da proteine. Così il giovane Perutz imparò le tecniche di cristallografia per applicarle alla biochimica. E scelse di studiare l'emoglobina, la proteina più abbondante e facile a cristallizzare, utilizzando quella estratta dal cavallo. Nel 1959 la struttura spaziale dell'emoglobina e della mioglobina era cosa fatta. Racconterà Perutz: "Eravamo come esploratori alla scoperta di un nuovo continente. Ma ora bisognava spiegare il meccanismo molecolare dello scambio respiratorio". L'obiettivo venne centrato da Perutz e Kendrew negli anni successivi al Nobel. Un meccanismo che si rivelerà la chiave per comprendere numerose malattie ereditarie. A cominciare dall'anemia falciforme, dovuta a una sintesi difettosa dell'emoglobina. Perutz aus Cambridge (England) wird am Donnerstag, 19. November, im Rahmen der "Rudolf Virchow Vortragsreihe" der Medizinischen Fakultät der Universität Würzburg sprechen. Mit dieser Vortragsreihe will die Fakultät hochkarätigen Wissenschaftlern ein Forum bieten und die Referenten gleichzeitig mit ihrer Auswahl ehren. Max F. Perutz kommt unter anderem das Verdienst zu, die Raumstruktur des roten Blutfarbstoffs Hämoglobin aufgeklärt zu haben. Dies war in der zweiten Hälfte des 20. Jahrhunderts einer der entscheidenden wissenschaftlichen Erfolge und wurde 1962 mit dem Nobelpreis für Chemie bedacht, den Perutz zusammen mit John Kendrew erhielt. Die Entdeckung von Perutz bereicherte nicht nur das Wissen über die Struktur der Proteine und ihre Regulation. Sie hatte auch großen Wert für die Medizin: Erstmals konnte molekular erklärt werden, warum die vielen abnormen Varianten des roten Blutfarbstoffs die normalen Lebensvorgänge stören.

Kauffman, California State University, Fresno george_kauffman@csufresno.edu As frequent book reviewers, we particularly enjoyed this fascinating collection of essays (22 of which are book reviews) by Max Ferdinand Perutz, the Austrian-born British biochemist who shared the 1962 Nobel Prize for Chemistry with John C. Kendrew for X-ray diffraction analysis of the structure of hemoglobin. In his preface to this well-chosen selection he describes hemoglobin as his “mistress.” The reviews transcend the genre and can be read with pleasure as independent essays. The book is suitable for both browsing and for careful reading Perutz was Director of the Medical Research Council Laboratory for Molecular Biology, Cambridge, England from its establishment in 1962 until 1979, and he remains a scientific staff member. To date nine MRC-LMB staff members have received ten Nobel prizes, and Perutz muses on the factors underlying such creativity. He concludes that:
