The physiology of insulin and its source in the animal body
The knowledge that the isles of Langerhans of the pancreas have the function of secreting into the blood a hormone which plays an essential role in the regulation of the metabolism of the carbohydrates, is the outcome of numerous investigations extending over many years, and to the development of this knowledge workers in various fields of medical science have contributed. The insular hypothesis of diabetes was meanwhile strongly supported by the careful histological studies of the pancreas of patients who had died from the disease (Opie) for, although it had been known, even prior to the experiments of Minkowski and von Mering, that the gland is often the seat of morbid change, it was not realized that the islets are the structures which are chiefly affected. In 1903-1904, Rennie, by anatomical studies in certain Teleostei, gave strong support to the view of Diamare, that the islet cells in these fishes exist as separate glands of relatively large size and more or less independent of the pancreatic acini. Both workers attempted to demonstrate an effect of extracts of these glands on sugar or starch solutions, but without success. They administered them by mouth to diabetic patients with no favourable results, although in one case, in which an extract was given subcutaneously, there was decided alleviation of the diabetic symptoms (Rennie and Fraser).

En 1898 se graduó, con honores, en Medicina y obtuvo una beca para trabajar durante un año en el Instituto de Fisiología de la Universidad de Leipzig. Tras seguir estudios de fisiología y bioquímica en el London Hospital Medical School fue nombrado, ya en 1903, profesor de Fisiología en la Westem Reserve University de Cleveland, Ohio, en Estados Unidos. En 1918 fue elegido profesor de esta misma disciplina en la Universidad de Toronto, Canadá. Allí fue también nombrado director del laboratorio de Fisiología y, asimismo, decano asociado de la Facultad de Medicina. En 1928 alcanzó el título de profesor regio de Fisiología de Universidad de Aberdeen y fue consultant physiologist del Instituto Rowett de Nutrición Animal cargo que ocupó hasta su fallecimiento, a la edad de cincuenta y nueve años. Desde 1905, MacLeod se centró en la investigación del metabolismo de los hidratos de carbono y la diabetes y publicó hasta un total de medio centenar de estudios sobre estos temas y sobre la glucosuria producida de forma experimental, siguiendo las investigaciones de Von Mering y Minkowski. † 31. März 1978 in Toronto Die Entwicklung des Insulins gehört zu den letzen abenteuerlichen und großen Erfolgsgeschichten der modernen Medizin, die zu Beginn der zwanziger Jahre stattgefunden hat. Innerhalb von nur zwei Jahren entdeckten vier Forscher das Insulin und entwickelten es weiter bis zur medizinischen Anwendung am Menschen. Die Lösungsansätze hat ohne Zweifel der kanadische Arzt Frederick Banting durch seine Überlegungen geschafften. Sein Mitarbeiter Charles Herbert Best war damals noch ein Student. Die Zuckerkrankheit kennt man schon seit dem Altertum. Die Kenntnis der Ursachen und erfolgreiche Behandlungsmethoden blieben der Forschung vorerst versagt. Die Empfehlung einer Diät war die gängigste Behandlungsmethode. Namhafte Physiologen hatten sich im 19. Jahrhundert intensiv um die Erforschung von Diabetes bemüht, und es wurden zahlreiche Versuche seit 1890 angestellt, um brauchbare Pankreasextrakte zur Behandlung zu gewinnen. Es war daher sehr überraschend, als das im Vergleich zu bekannten Forscherexperten und Forscherteams unerfahrene Forscherduo Banting und der studentische Best in weniger als einem Jahr das Hauptproblem der Insulinisolierung lösen und die Anwendung im Humanexperiment auch tatsächlich erfolgreich beschließen konnte.

Berliiniläinen Georg Ludwig Zülzer onnistuikin eristämään vuonna 1903 tehokkaan valmisteen. Sen riittämätön puhdistaminen ei kuitenkaan tehnyt mahdolliseksi kokeita ihmisillä. Farmaripariskunta William Thompson ja Sarah Grant Banting saivat vuonna 1891 Bonhead'issa Kanadan Ontariossa pojan, joka sai nimen Frederick Grant Banting.
