Otto Loewi
Born in Frankfurt am Main, Germany, Otto Loewi originally wanted to be an art historian but bowed to family pressures and went to medical school instead (similarly, Ramón y Cajal's plans to become a painter were thwarted by his father). Loewi later went into pharmacology research and was one of the first people to propose that cells talked to one another with "little whiffs of scent" - more formally to be known as the chemical theory of synaptic transmission. While living in Graz, Austria in 1921, Loewi dreamed of an experiment that would win him the Nobel Prize - "The night before Easter Sunday of that year I awoke, turned on the light, and jotted down a few notes on a tiny slip of thin paper. Then I felt asleep again. It occurred to me at six o'clock in the morning that I had written down something most important, but I was unable to decipher the scrawl. The next night, at three o'clock, the idea returned. It was the experiment to determine whether or not the hypothesis of chemical transmission that I had uttered seventeen years ago was correct.     Otto Loewi was born on June 3, 1873, in Frankfurt-am-Main, Germany. He was torn between medicine and philosophy but by 1894 he became dedicated to the idea of being a doctor. By 1896 he got his doctorate and after seeing the high toll tuberculosis was causing, he decided to go into research to work on a cure. He discovered many essential ideas in biochemistry, including carbohydrate metabolism. In the early 20s, he was working on how nerves transmit impulses. He worked on it night and day with little result. Then one night he fell asleep and had a vivid dream. He scrawled down some notes but was unable to read them the next morning. Frustrated, he waited until the next night. Again, he had a vivid dream, showing him the style of experiment that would help him in his nerve transmission work. Sure enough, he went immediately to his lab to try the experiment. It worked, and as a result, Otto Loewi was awarded the 1936 Nobel Prize for Medicine.

    The chemical transmission of nerve action
    Natural or artificial stimulation of nerves gives rise to a process of progressive excitation in them, leading to a response in the effector organ of the nerves concerned. Up until the year 1921 it was not known how the stimulation of a nerve influenced the effector organ's function, in other words, in what way the stimulation was transmitted to the effector organ from the nerve-ending. In general it was thought that it came about through direct transmission of the stimulation wave from the nerve fibre to the effector organ. But the possibility of transmission by chemical means had also been considered and experiments had been conducted on these lines. As a result of his own experiments, Howell had come to believe that vagus stimulation released potassium in the heart and that this was the cause of the resultant effect, and Bayliss discussed the possibility, in view of the similarity in action of the so-called vagomimetic substances and chorda stimulation, that this stimulation might be caused by the production of such substances.     Otto Loewi's Frog Heart Experiments The Problem: In 1903 Otto Loewi wondered why one of the heart nerves (the accelerator) speeds up the heart and the other (the vagus) slows it down, even though the electrical pulses of the nerves were nearly identical. The Hypothesis: Loewi suggested that the 2 nerves released different chemicals at their terminals when stimulated. The vagus chemical would slow down the heart and the accelerator chemical would speed it up. The Experimental Test Seventeen years later Loewi had a very strange dream in which a way of testing his hypothesis appeared. He woke up and wrote down some notes, but the next morning he could not read his scribbles! Luckily for him the dream reoccurred the next night and he immediately got up (3AM) and did the experiment. Hearts, with their nerves intact, were dissected from 2 frogs. The hearts were hooked up to mechanical devices which recorded their heart beats. Then the vagus nerve of one heart (the donor) was electrically stimulated. The donor heart immediately slowed down, as expected. A sample of the fluid passing through the donor heart was taken with a small pipette and this fluid was dripped onto the second heart (the recipient). The recipient heart slowed down, even though its vagus had not been stimulated. Chemicals released by the donor heart into the fluid were sufficient to produce slowing. (Note: the picture shows one of several ways of doing this experiment. In some of the early experiments the fluid from the donor heart was collected in a beaker and the recipient heart was placed directly into the beaker).
