Nobel Laureate Born and raised in Brooklyn, New York, Dr. Kornberg is a 1941 graduate of the University of Rochester's School of Medicine and Dentistry. After serving in the U.S. Navy during World War II, he returned to conduct research at the National Institutes of Health, NYU, Berkeley, and Washington University. Soon after he joined the faculty of Stanford University in 1959, he received the Nobel Prize in Medicine (with Dr. Severo Ochoa of New York University) for their discoveries of the mechanisms of the biologic synthesis of RNA and DNA. Dr. Kornberg has made significant contributions to our understanding of the mechanisms of inheritance and to the development of modern recombinant DNA technology. The Arthur Kornberg Research Awards were established in 1997 to recognize faculty in the School of Medicine and Dentistry for excellence in biomedical research.

Arthur Kornberg Medical Research Building
recruitment of 50 additional scientists (including several of the world's foremost authorities in various fields) and erected a state-of-the-art, 225,000 square-foot research building on the Medical Center campus. By concentrating its resources on these six fields, the Medical Center will assemble a scientific powerhouse in each discipline that, with the exception of its research program in Cancer Biology, will be virtually unmatched in any other institution. Research laboratories in the Aab Institute of Biomedical Sciences feature state-of-the-art scientific equipment. The architectural signature of the new research building is the dramatic point on the west side, which houses the triangular break rooms on each floor. The purpose of these rooms is to foster interaction and the exchange of ideas among scientists in an informal, relaxed setting.

Numerous and varied biological functions are performed by poly P depending on the need and where it is needed-the species, cell, and subcellular compartment. Among these functions are: reservoirs of energy and phosphate, chelation of metals (e.g., Mn , Ca ), buffer against alkali, capsule of bacteria, competence for bacterial transformation, ecological disposal of pollutant phosphate, and, of great interest, physiologic adjustments to growth, development, stress and deprivation. Our approach has been to discover the enzymes for the synthesis and utilization of poly P and when purified open the route of reverse genetics: the peptide sequence leads to the gene and thereby the means to knock it out and overexpress it. By manipulating expression of the gene and the cellular levels of its product, phenotypes are created which provide clues to metabolic functions. Most immediate and decisive, the enzymes provide unique and invaluable reagents for analytic and preparative work.

Specific Information on a Specific Contribution National Institutes of Health (1942-1953), he helped elucidate the reactions leading to the formation of two important coenzymes: flavin adenine dinucleotide (FAD) and diphosphopyridine nucleotide (DPN). D. Conclusion Professor Kornberg finds time to travel and lecture at many universities and research centers. He calls for a return of simple curiosity. He lectures that “ scientists need to be able to study science for curiosity’s sake and not be driven by the possible dollar benefits-and governments should encourage them.” He is adamant that research into science should proceed, despite public concerns that new discoveries, especially in genetic engineering, could be abused. “There is often a focus on the alarmist possibilities of any new technology; however it is foolish to try and predict where science will take us.” He states “ there hasn’t been a single instance of biotechnology being misapplied. It’s safer than driving.” Professor Kornberg is a remarkable individual, and his devotion to his field of study is inspirational. His contributions have advanced medical knowledge. Medical knowledge needs to constantly advance because of the challenges of new problems- such as novel toxins and resistant organisms that are constantly arising. I believe that research is the lifeline to medicine; we should continue to question and search for answers.

    From his early studies of the mechanisms of the enzymatic synthesis of coenzymes and inorganic pyrophosphate, he extended his interest to the biosynthesis of the nucleic acids, particularly DNA. After elucidating key steps in the pathways of pyrimidine and purine nucleotide synthesis, including the discovery of PRPP as an intermediate, he found the enzyme that assembles the building blocks into DNA, named DNA polymerase. This ubiquitous class of enzymes make genetically precise DNA and are essential in the replication, repair and rearrangements of DNA. Many other enzymes of DNA metabolism were discovered responsible for the start and elongation of DNA chains and chromosomes. These enzymes were the basis of discovery of recombinant DNA which helped ignite the biotechnology revolution. Since 1991, he switched his research focus from DNA replication to inorganic polyphosphate (poly P), a polymer of phosphates that likely participated in prebiotic evolution and is now found in every bacterial, plant and animal cell. Neglected and long regarded a molecular fossil, he has found a variety of significant functions for poly P that include responses to stresses and stringencies and factors responsible for motility and virulence in some of the major pathogens.

    Biochemist Arthur Kornberg: A Lifelong Love Affair With Enzymes
    By [Editor’s note: While best known for his discovery of DNA polymerase and his subsequent synthesis of DNA molecules, Nobel Prize-winning biochemist Arthur Kornberg considers himself first and foremost a researcher of enzymes. Over the course of his career, which now spans a half-century, he has continued to pursue the study of these catalytics as other biochemist s—much to his chagrin—have chosen to ignore them. “Without knowing and respecting enzymes, better still loving them, answers to the most basic questions of growth, development, and disease will remain beyond reach,” he has said. Today, at the age of 71, Kornberg continues his enzyme research at Stanford University, where he has been a professor and chairman of the biochemistry department. He also continues to lecture; when contacted by The Scientist at Stanford !ast month, he was about to depart for Seoul, South Korea, where he was to make a presentation to the Congress of the Federation of Asian and Oceanian Biochemists.
