Paul Greengard received his Ph.D. from Johns Hopkins University in 1953. Afterward, he spent five years in England where he received advanced training at the University of London, University of Cambridge and National Institute for Medical Research. Dr. Greengard then worked as the department of biochemistry's director at Geigy Research Laboratories in New York, after which he served as professor of pharmacology and psychiatry at Yale University School of Medicine.
Dr. Greengard's achievements have earned him many distinguished awards and memberships in prestigious societies. In 2000, Dr. Greengard was awarded the Nobel Prize in Physiology or Medicine for his discoveries, concerning signal transduction in the nervous system. In the course of his career, he has authored over 1,000 scientific publications.
Today, he is Vincent Astor Professor and Head of the Laboratory of Molecular and Cellular Neuroscience at The Rockefeller University and Director of the Zachary and Elizabeth M. Fisher Center for Research on Alzheimer's Disease at Rockefeller.

PAUL GREENGARD, TO ITS SCIENTIFIC ADVISORY BOARD Dr. Greengard's discoveries provide a conceptual framework for understanding how the nervous system works at a molecular level. His research has demonstrated that the effects of therapeutic and toxic drugs can be explained through distinct neurochemical actions which affect the transmission of nerve signals in the brain. Dr. Greengard's research is directed toward understanding the signal transduction mechanisms that underlie communication between neurons in health and disease. His current studies include characterization of the role of the synapsins, the most abundant brain phosphoproteins, in regulation of neurotransmitter release and synaptogenesis, determination of the mechanisms underlying interactions of dopaminergic and other signal transduction pathways involved in schizophrenia, Parkinson's disease and drug abuse, and elucidation of the mechanisms by which protein phosphorylation regulates the formation of the b-amyloid responsible for Alzheimer's disease.

Több évtizedes kutatásaik nemcsak az agymûködés jobb megértéséhez vezettek, de a Parkinson-kór, az Alzheimer-kór, a skizofrénia és a depresszió kezelésére alkalmas új gyógyszereket is kifejlesztettek az õ kísérleti eredményeik alapján. A három tudós december 10-én megosztva kapja meg a kilencmillió svéd koronával (276 millió forint) járó díjat.

A 77 éves svéd Arvid Carlsson a Göteborgi Egyetem gyógyszerészeti tanszékét vezeti. A 74 éves Paul Greengard a New York-i Rockefeller Egyetem sejt- és molekuláris neurológiai laboratóriumának kutatója, a 70 éves, osztrák születésû amerikai állampolgár, Eric Kandel pedig a New York-i Columbia Egyetem neurobiológiai és viselkedéstudományi központjában dolgozik. Mindhárom tudós az idegsejtek közti jelátvitel egyik altípusa, az úgynevezett lassú szinaptikus ingerületátvitel kutatásán dolgozott. Több évtizedes munkájuk nemcsak az agymûködés jobb megértéséhez vezetett, de új gyógyszereket is kifejlesztettek az õ kísérleti eredményeik alapján. Nélkülük nem lenne Prozac Arvid Carlsson és Paul Greengard a dopamin nevû ingerület-átvivõ anyag viselkedését kutatták. Arvic Carlsson volt az elsõ, aki az ötvenes években felfedezte, hogy a dopamin fontos szerepet játszik az agyi ingerület-átvitelben és a mozgáskoordinációban. Kutatásai során rájött, hogy a Parkinson-kórt a dopamin hiánya okozza, és hogy az L-dopa vagy levadopa nevû anyag adagolásával elkerülhetõ a betegség súlyosbodása. Carlsson kutatócsoportja a hatvanas években kifejlesztette a depresszió ellen alkalmazható elsõ klinikailag aktív szerotonin-felvételt blokkoló anyagot, a zimelidint. Kutatásai alapján született meg késõbb a hasonló elven mûködõ Prozac, az egyik legnépszerûbb antidepresszáns.

Research Interests: Through yeast two-hybrid screening, we identified a number of proteins that may potentially interact with parkin. Base on one of the leads, we found that parkin bound to a b tubulin and microtubules very strongly and increased the ubiquitination and degradation of a - and b -tubulins. It has been shown that PD-causing neurotoxins such as rotenone and MPP strongly depolymerize microtubules. As microtubules play obligatory functions in the maintenance of cellular morphology and intracellular transport, their depolymerization by these toxins would cause significant damages to the cell, especially for long projection neurons such as dopamine cells on the nigrostriatal pathway. Among the many things that could be affected by microtubule depolymerization, lack of sufficient transport of dopamine vesicles to the terminal would directly reduce dopamine release in the striatum. Thus, the microtubule-depolymerizing activity of MPP and rotenone may significantly affect the normal functions of nigral dopaminergic neurons. Parkin, through its ability to ubiquitinate and degrade misfolded tubulin, may protect neurons from toxic accumulation of misfolded tubulin, particularly when the cells are exposed to neurotoxins such as MPP
