Signal Transduction in the Brain
Presented on August 10, 2001 by Paul Greengard, PhD, Laboratory of Molecular and Cellular Neuroscience, The Rockefeller University
"...in a way, signaling in the brain is complicated, and yet on the other hand [it is] beautifully simple. The more we learn, the more complex these pathways are that we have been working on, the more relatively simple principles we find."
Paul Greengard
Paul Greengard, Ph.D., Vincent Astor Professor of Molecular and Cellular Neurosciences at Rockefeller University and winner of the 2000 Nobel Prize in Physiology or Medicine for his work in communication between nerve cells, talks about current understanding of the complex biochemistry of signal transduction in the brain. He discusses the implications of his ongoing work in the potential for novel pharmaceutical treatments of various neurological and psychiatric disorders.
Paul Greengard, Ph.D., Vincent Astor Professor of Molecular and Cellular Neurosciences at Rockefeller University, has devoted most of his research career to studying communication between nerve cells. His contribution to our understanding of biochemical signaling in the brain was recently recognized with the award of the 2000 Nobel Prize in Physiology or Medicine.

Signal Transduction in the Brain: Transcript: Introducing Paul Greengard
Introduction by George Augustine:
My name is George Augustine and I am here to introduce tonight's Friday evening lecturer, Dr. Paul Greengard from the Rockefeller University. Paul is a great scientist and a great friend and it is a genuine honor to introduce him to you.
Paul studies signaling processes in the brain, with a special interest in synaptic transmission. As most of you probably know, the power of the brain resides in the very sophisticated abilities of nerve cells to communicate with each other. Paul's work has brought our appreciation of these synaptic communication processes to an unprecedented level of understanding. Because he has published more than 1,000 papers on this subject, it is impossible for my introduction to do justice to his interests or to his contributions. I expect that his talk with offer you an excellent overview of this work.
By way of introduction I think it is fair for me to say that Paul's main contribution has been to vastly expand our views of synaptic signaling. When Paul began his work, our view of synaptic transmission largely started and ended with the electrical signals that are generated at synapses. While these electrical signals are undeniably important, Paul's work has shown that a far more elaborate cascade of chemical signaling events occurs within nerve cells. These signaling processes allow very sophisticated chemical computations to occur within the nerve cells and these chemical computations help sculpt the electrical signals that are ultimately produced by the nerve cells.

PAUL GREENGARD

The Nobel Prize in Physiology or Medicine This year, the Nobel committee awarded the Prize for Physiology or Medicine to three neuroscientists, Arvid Carlsson, Paul Greengard, and Eric R. Kandel. Their contributions opened a molecular window onto the brainCarlsson by identifying dopamine as the first brain neurotransmitter found uniquely in the brain, Greengard by illuminating the molecular cascade triggered inside the neuron by dopamine, and Kandel by recognizing that the molecular basis of cognitive processes such as learning must first be worked out in simpler systems before they can be understood in the brains of complex mammals such as ourselves. Science has had the privilege of publishing many of the seminal papers from two of these scientists, Paul Greengard and Eric R. Kandel. A selection of some of the most important follows:

2000 Nobel Laureate
Director of the Fisher Center for Alzheimer's Research at The Rockefeller University
When Paul Greengard was a graduate student in the late 1940s, the question of how the brain's chemical messengers produce their effect on nerve cells was so deeply mysterious and seemed so impossible to answer that most scientists largely avoided asking it. But the young Greengard dared to ask the question, and spent most of the next 50 years unraveling the answers. In the process, he has spawned an entire new branch of neurobiological research, has revolutionized our understanding of the biochemical mechanisms involved in communication between nerve cells and has forged a path to novel treatments for brain disorders such as Alzheimer's disease. Dr. Greengard's pioneering work in delineating how neurons communicate with one another in the brain earned him the 2000 Nobel Prize in Physiology or Medicine. During a half-century of research, he has arguably contributed more than any other single scientist to our understanding of the complex signaling processes that occur within each of the 100 billion or more nerve cells in the human brain. Collectively, these cells orchestrate every aspect of our being - from walking and talking to thinking and feeling emotion to having a unique personality. Understanding these processes enables us to comprehend not only the fundamental nature of brain function, but also what goes wrong in brain diseases such as Alzheimer's and Parkinson's. The ultimate goal, of course, is to develop effective treatments for these devastating disorders.

The Fisher Center Foundation is sponsoring a documentary entitled "Alzheimer's - Is There Hope?" The hour long exploration of the tragedy of Alzheimer's disease covers the disease from its origins to recent breakthroughs that give sufferers and caregivers reason for optimism. The program is to air on public television stations beginning November 2002. The documentary is to coincide with the official launch of the Foundation's Alzheimer's information website, Alzinfo.Org. The documentary is being produced by WLIW, Channel 21 in New York. As its title suggests, the documentary will investigate the hope that currently exists in the battle against Alzheimer's disease. The documentary will feature the leading experts in the field - including Fisher Center Director Dr. Paul Greengard who discuss the latest research gains, improvements in drug regimens, and other encouraging developments. Families and caregivers who have been affected by the disease will also be featured on the documentary. For more information about the documentary
