Paul Greengard . Född i New York City . Amerikansk Nobelpristagare i fysiologi eller medicin år . Han tilldelades priset för sina " upptäckter rörande signalsubstanser i nervsystemet ". Han delade priset med sin landsman Eric R Kandel och svensken Arvid Carlsson
Signalerna från en nervcell till en annan överförs med hjälp av olika signalsubstanser . Signalöverföringen sker i speciella kontaktpunkter, synapser . En enda nervcell kan ha tusentals kontaktpunkter med andra nervceller. Arvid Carlsson, har upptäckt att dopamin är en sådan signalsubstans i hjärnan och att dopamin har stor betydelse för kontroll av våra rörelser. Hans forskningsrön ledde i sin tur till insikten att Parkinsons sjukdom orsakas av dopaminbrist i vissa delar av hjärnan och till att man kunde få fram ett effektivt läkemedel (L-dopa) mot denna sjukdom. Paul Greengard, upptäckte hur dopamin och en rad andra signalsubstanser utövar sina effekter i nervsystemet. Signalsubstanserna påverkar först en receptor på cellens yta. Det utlöser en kaskad av reaktioner som påverkar vissa "nyckelproteiner" som i sin tur reglerar olika funktioner i cellen. Proteinernas form och funktion förändras genom att fosfatgrupper tillförs (fosforylering) eller tas bort (defosforylering). Genom denna mekanism kan signalsubstanser överföra sina budskap mellan nervceller. Eric Kandel,upptäckte sedan hur synapsernas effektivitet kan förändras och med vilka molekylära mekanismer detta sker. Han visade att synapsernas funktion är central för inlärning och minne. För uppkomsten av en form av korttidsminne spelar proteinfosforylering i synapsen en viktig roll. För att ett långtidsminne ska uppstå krävs dessutom nybildning av proteiner som bl.a. leder till att synapsens form och funktion förändras.

My paternal great, great grandfather emigrated from Koenigsberg, now Gdansk, to St. Louis during the 1850s, a time of large-scale movement of Germans, including German Jews, to the Mid-Western United States. My paternal grandfather moved to Binghamton, New York, where my father was born. My father had success in vaudeville as a singer/dancer/comedian. He eventually became a businessman working first in retail and then in wholesale in the cosmetics field. I attended public schools in Brooklyn and Queens. During World War II, I spent three years in the Navy as an electronics technician. After appropriate training, I was assigned to a team at the Massachusetts Institute of Technology that was involved in developing an early-warning system to intercept Japanese kamikaze planes before they could reach the ships of the U.S. fleet. After the war, I attended Hamilton College , a small liberal arts college located in Clinton, New York, where I majored in mathematics and physics, and from which I graduated in 1948. I had been interested in going to graduate school in theoretical physics, but decided not to do so because at that time the only fellowship support for such graduate studies came from the Atomic Energy Commission. This was only three years after dropping the atomic bombs on Japan, and I didn't want to contribute to research the fruits of which might contribute to creating more powerful weapons of mass destruction. In thinking about various options, I settled on the then nascent field of biophysics. At that time there were two groups of academic biophysicists. One, at the University of California, was engaged in biological and medical applications of radioisotopes. The other, at the

Vincent Astor Professor The goal of our laboratory is to understand more fully the molecular basis of communication between neurons in the mammalian brain. Toward this end, we have developed a general model of signal transduction in the nervous system. The major molecular mechanism underlying signal transduction is protein phosphorylation. Synaptic Vesicle-associated Phosphoproteins. The major synaptic vesicle-associated phosphoproteins that we have identified and characterized are synapsin I, synapsin II, and synaptophysin. As an example of the functioning of this class of phosphoproteins, we have obtained evidence that dephosphorylated synapsin I functions to tether synaptic vesicles in a "reserve" pool. When synapsin I is phosphorylated, this tethering function is abolished, and the synaptic vesicles are then free to enter a "readily releasable" pool of synaptic vesicles. Additionally, we have obtained evidence that the synapsins are involved in the formation of new synapses. Neostriatum-enriched Phosphoproteins.

Paul Greengard, one of this year’s winners of the Nobel Prize in medicine, joins Dr. Steve Hyman, director of the National Institute of Mental Health, to talk about his research and winning the Nobel Prize. RAY SUAREZ: The award this year went for discoveries about how messages move around the nervous system and help us understand the functioning of the brain. Three scientists will share the nearly $1 million prize. One of them is with us now. Paul Greengard is a neuroscientist who heads the laboratory of molecular and cellular neuroscience at the Rockefeller University in New York. Also joining us is Dr. Steve Hyman, director of the National Institute of Mental Health.
First, Professor Greengard, congratulations. PAUL GREENGARD: Thank you very much. RAY SUAREZ: How did you hear about the award? PAUL GREENGARD: About ten minutes past five this morning, the secretary of the Nobel Committee called to inform me I had been one of the recipients of this year's Nobel Prize in medicine. It was about 20 minutes... apparently in Sweden they announce at 11:30 in the morning, which is 5:30 Eastern Standard time. So they called to let the recipients know in advance of the deluge of telephone calls that come after the announcement of the prize.

LONDON. So, it wasn't the human genome; nor the nematode. Today, nerves seem to have got to the Nobel Assembly at the Karolinska Institute, Stockholm, which this morning announced the award of the Nobel Prize in Physiology or Medicine for 2000 jointly to Arvid Carlsson, Paul Greengard and Eric Kandel. In the 1990s he extended these studies to mice, and showed that the same type of changes in synaptic function also take place in mammals. This Nobel is Kandel's 29th scientific prize or medal, including the Lasker Award (with VB Mountcastle) in 1983. Arvid Carlsson, 77, of the Department of Pharmacology, University of Gothenburg, was rewarded "for his discovery that dopamine is a transmitter in the brain and that it has great importance for our ability to control movements." The Karolinska emphasises Carlsson's contributions to the development of L-dopa as a treatment for Parkinson's disease, to the understanding of the action of drugs for schizophrenia, and the development of selective serotonin uptake blockers as antidepressants. My paternal grandfather moved to Binghamton, New York, where my father was born. My father had success in vaudeville as a singer/dancer/comedian. He eventually became a businessman working first in retail and then in wholesale in the cosmetics field.
I was born on December 11, 1925 in New York City under tragic circumstances - my mother, née Pearl Meister, died giving birth to me. My father remarried when I was 13 months old. In contrast to my mother, who had been Jewish, my stepmother, who raised me, was Episcopalian. From that time on I was brought up in the Christian tradition, celebrating Easter, Christmas, etc. I was prevented access to my biological mother's family with whom I became familiar only very recently. I have been delighted to learn that many members of that family are highly creative individuals working in various fields of science, government, etc.
