Molemmat löydöt ansaitsivat varmasti artikkelissa paikkansa eli palkitsemisen myöntämisperustelua lainaten: "for their discoveries concerning new mechanisms for the origin and dissemination of infectious diseases". Kuru Culture Donald J. McGraw The Trembling Mountain: A Personal Account of Kuru, Cannibals and Mad Cow Disease. Robert Klitzman. Plenum, 1998. $27.95. Every word in the subtitle is an irresistible lure: The promise, however, is only partially fulfilled. This book is very much a personal account of kuru and of its mechanism of spread, cannibalism. But there are only a few pages about bovine spongiform encephalopathy, or mad cow disease. That may easily be explained in two ways: There is yet little that can be said about this newest of the prion diseases, and this is not a book of science per se. Robert Klitzman's personal account is not meant to satisfy the biological scientist with new information on any aspect of the subtitle except to convey his discoveries in the field on certain aspects of kuru, or the "trembling," in the language of the Papua New Guinea tribe for whom the disease has been and still may be rampant. The Fore, as highlands cannibals, have long been victims of this frightening malady, and Klitzman tells of his findings in kuru's transmission, symptomology and the primary orientation of this account, its sociology.

Scientists in half a dozen countries have been vying to work with Nobel laureate D. Carleton Gajdusek, who was released from prison this week after serving a year on charges of child sexual abuse. Gajdusek immediately jetted off for the Continent, where he has enough offers to keep him busy indefinitely. Gajdusek shared the Nobel Prize in physiology or medicine in 1976 for his groundbreaking research into kuru, a brain disease similar to bovine spongiform encephalopathy that affected tribes in New Guinea. In 1996, the virologist was charged with sexually abusing a teenaged Micronesian boy living in his home ( Science , 12 April 1996, p. 203). After pleading guilty in February 1997 as part of a plea bargain, Gajdusek resigned as chief of the National Institutes of Health's Laboratory of Central Nervous System Studies and was later sentenced to 18 months in prison. Now that he's free, Gajdusek "wishes to roll around among different labs like Diogenes in a barrel, as he expressed it," says Georg Klein of the Karolinska Institute in Stockholm, where Gajdusek is expected to show up in July. Then, in September, he will begin a 6-month visiting professorship at the Department of Human Retrovirology of the Academic Medical Center in Amsterdam, arranged by virologist and AIDS researcher Jaap Goudsmit. Says Goudsmit, who worked as a postdoc in Gajdusek's lab from 1979 to 1981, "I'm very fond of him. He is still very smart and he knows a lot about viruses. It will be wonderful to have him."

    D. Carleton Gajdusek, winner of the 1976 Nobel Prize in medicine, was invited by the Beijing Inspection and Quarantine Bureau to act as a consultant for the bureau's mad cow disease lab.
Gajdusek, awarded the Nobel Prize for his discoveries concerning "new mechanisms for the origin and dissemination of infectious disease," said he would help China improve its researchand prevention of mad cow disease.
Wei Chuanzhong, director of the bureau, said the issue of food safety has increasingly become a concern for all governments and the spread of mad cow disease poses a threat to mankind.
He said he believes that with the American laureate's expertise, the lab will advance in terms of testing methods for the disease.
The bureau's mad cow disease lab is China's first and has mastered a number of ways to test for the disease. A research center is in the planning to further the country's fight against the deadly disease.

By Multiple Concerns over governance have nixed a merger between New York's Mount Sinai and New York University medical centers. The proposed venture, announced in June ( M.E. Watanabe, The Scientist , Aug. 19, 1996, page 1 ), would have united the hospitals and medical schools of the two institutions. But according to a report in the February 15 New York Times (E.B. Fein, page 25), the deal fell through because Mount Sinai and NYU could not agree on who would control the combined medical school. Sources told the Times that Mount Sinai saw the school as "an independent entity involving equal partnership between the two sides," while NYU "wanted the medical school to be an integral part of its university, like its law school, film school, and other divisions." Neither John W. Rowe, Mount Sinai's president, nor NYU dean Saul Farber returned calls from The Scientist seeking comment. In a joint statement, the institutions noted that they "could not find the appropriate way to combine the academic components." However, they expressed hope that they could "continue the many areas of collaboration that pre-existed our talks, as well as others we discovered in the process." Photo: Lynn Gambale

Figure 1. The multidimensional sequence space defines all possible sequences of amino acids. Among all possible sequences nature selected only those (about 350,000 sequences total) that fold and carry certain functions. How big is the fraction of amyloidogenic proteins and how many proteins possess specific features of prions are questions of fundamental interest. Research in my lab is focused on the molecular mechanism of the conformational transitions of the prion and non-prion proteins and address basic issues of protein folding. They include the possibility of forming distinct long-lived states within the same primary structure, the position of the native state versus the abnormal states in the energetic landscape, and the overall complexity of the energy landscape of folding. Our previous studies revealed that the folding of the prion protein (PrP) to its native a-helical conformation is under kinetic rather than thermodynamic control (Figure2). We found that abnormal b-sheet rich isoform is thermodynamically more stable than the native a-helical isoform. The conformational transition from the a-PrP to the b-PrP is separated by a large energetic barrier that is associated with unfolding and with a higher order kinetic process of oligomerization. Although the b-PrP is thermodynamically more stable than the a-PrP, the process of conformational transition is prevented over the protein's lifetime because of high energetic barrier. Thus, to avoid formation of non-native forms and to optimize the efficacy of the native folding pathway, thermodynamic and kinetic parameters should be subjects of natural evolution.
