Gerhard Domagk Gerhard Domagk was born in 1895 in Lagow, Brandenburg, as the son of a teacher. At the age of 20, Domagk - who would later go on to win a Nobel Prize - was confronted with the supposed limits of medicine through his experiences in World War I. At that time, operations that were initially successful were often marred by fatal infectious diseases such as gangrene or gas gangrene. The antiseptic properties of the then-common substances chlorine water and carbolic acid did not last long enough to effectively combat these diseases. In 1914 Domagk received his diploma from the upper secondary school in Liegnitz and began a course of study in human medicine at the University of Kiel in the same year. After having to interrupt his studies for a time due to the war, he received his doctorate in 1921. His post-doctoral qualification was obtained in 1924 at the Pathological Institute of the University of Greifswald. In 1927 the attention of Professor Heinrich Hoerlein was drawn to Domagk's post-doctoral thesis entitled "Destroying infectious diseases through the reticuloendothelium and the development of amyloid." He recruited the young scientist to the pharmaceutical department in Elberfeld. In 1932, while searching for a way to combat bacterial infections by chemical means, Domagk stumbled upon the active substance dimethyl benzyl dodecyl ammonium chloride, which was later launched on the market as a 10 percent solution under the trade name Zephirol. Zephirol's intensive antibacterial effect and good skin compatibility quickly made the product an indispensable and universally applicable skin disinfectant that is still used today for disinfecting hands and instruments.

Domagk, Fleming, Waksman and the Third Man. What do these people have in common and who is the third man?
The Nobel Prize for Medicine has been awarded for discovery of three different antibiotics:
    • To Gerhard Domagk for prontosil rubrum in 1939.
    • To Alexander Fleming for the discovery of penicillin in 1945.
    • To Selman Waksman for the discovery of streptomycin in 1952.
Fleming is still famous. You may have heard of Waksman. Domagk is largely forgotten.
Want to know more about the discovery of these antibiotics ?
If you want to know what this music has to do with antibiotics, you'll have to read to the end of the tutorial!

you can see the structure of sulphanilamide here The active metabolite - sulphanilamide - was already being produced by the ton as a side product of the dye industry. The patent to make sulphanilamide was owned by Bayer but had expired. In 1939 Gerhard Domagk was offered the Nobel Prize for Medicine. What happened next? He was interrogated by the Gestapo for eleven days. In 1935 a German pacifist opposed to Hitler had been awarded the Nobel Prize for Peace. As a result Hitler had declared that awarding a Nobel Prize to a German would be considered an anti-German act. Gerhard Domagk also discovered the thiosemicarbazone group of antibacterials and isoniazid. He finally collected his Nobel Prize in 1947

He also started working at the IG Farben laboratories at Wuppertal . The same year, he married Gertrud Str¼be. Later they would have three sons and a daughter. At IG Farben, he continued the studies of Josef Klarer and Fritz Mietzsch, based on works by Paul Ehrlich , to use dye s, at that time a major product of IG Farben, as antibiotic s.
