Né pendant la Première Guerre mondiale près de Londres, où ses parents se sont réfugiés, Christian de Duve entre à l'UCL en 1934. Il se tourne vers la médecine en raison de la pénurie de médecins provoquée par la Grande Guerre. Docteur en médecine en 1941, il poursuit ses recherches à Louvain pendant la Seconde Guerre mondiale, en se faisant passer pour un simple étudiant. Sa seule préoccupation : la solution au mécanisme de l'action de l'insuline. Agrégé (1945) et licencié en sciences chimiques (1946) de l'UCL, il devient professeur dans cette même Université (1951) et au Rockefeller Institute de New York. Il enseigne la chimie physiologique, la biochimie et la biochimie cellulaire, de même que la cytologie biochimique. Son mérite est d'avoir pu, aidé par d'excellents collaborateurs, développer avec énormément de patience et d'ingéniosité, les techniques de centrifugation des broyats cellulaires et de purification des fractions ainsi séparées grâce à leurs différences de densité. Son équipe parvint aussi à montrer que les diverses structures visibles dans les cellules sont des entités fonctionnelles bien définies, limitées par une vraie membrane, à l'intérieur desquelles sont concentrées des enzymes spécifiques, véritables marqueurs de ces structures.

Christian de Duve Christian de Duve, born in 1917 in Thames-Ditton, Great Britain, from Belgian parents, embarked on his career as a researcher when he began his medical studies (1934-1941) at the Catholic University of Louvain where he was appointed Professor in 1947. Initially, he became involved in the physiology of endocrinology and rediscovered glucagon, the insulin antagonist hormone, and then became interested in metabolic biochemistry. From 1950, he devoted himself to subcellular biochemistry, a new field which he was to pioneer with Albert Claude and George Palade. He particularly concentrated on refining fractionation techniques to isolate the various subcellular compartments and to establish their corresponding contribution to the cell's economy. His work has led to the discovery of lysosomes and peroxysomes, intracellular organs containing enzymes essential to biochemical reactions. He divides his time between the Catholic University of Louvain and the Rockefeller Institute (now the Rockefeller University) where he was appointed Professor in 1962 and he is presently Professor of Biochemical Cytology Emeritus. Christian de Duve is steadfast in his pursuit of a twin objective: to gain a greater in-depth knowledge of fundamental biology and to apply the most recent discoveries to the improvement of man's well-being and health.

Christian de Duve is a Belgian cytologist and biochemist, born in 1917, who discovered lysosomes (the digestive organelles of the cell) and peroxisomes (organelles that are the site of metabolic processes involving hydrogen peroxide). For this work he shared the Nobel Prize in Physiology or Medicine with Albert Claude and George Palade in 1974. In the course of research on the action of insulin upon liver tissue in 1949, de Duve observed a delay in the action of enzymes that break down material, suggesting the enzymes were enclosed with a membranous envelope. He calculated the probable size of this organelle, christened it the lysosome and later identified it in electron microscope pictures. He refined the cell fractionation techniques of Claude to isolate the lysosomes and perixosomes for study. In 1947, de Duve joined the faculty of Leuven University where he had received his Masters Degree in chemistry in 1946. From 1962, he simultaneously headed research laboratories at the Universities of Leuven, Belgium and Rockefeller, New York City.

For this work he shared the Nobel Prize for Physiology or Medicine in 1974 with Albert Claude and George Palade.
De Duve's discovery of lysosomes arose out of his research on the enzymes involved in the metabolism of carbohydrates by the liver. While using Claude's technique of separating the components of cells by spinning them in a centrifuge, he noticed that the cells' release of an enzyme called acid phosphatase increased in proportion to the amount of damage done to the cells during centrifugation. De Duve reasoned that the acid phosphatase was enclosed within the cell in some kind of membranous envelope that formed a self-contained organelle. He calculated the probable size of this organelle, christened it the lysosome, and later identified it in electron microscope pictures. De Duve's discovery of lysosomes answered the question of how the powerful enzymes used by cells to digest nutrients are kept separate from other cell components. In 1947 de Duve joined the faculty of the Catholic University of Louvain, Belg., where he had received his M.D. in 1941 and a master's degree in chemistry in 1946. From 1962 he simultaneously headed research laboratories at Louvain, where he became emeritus professor in 1985, and at Rockefeller University, New York City, where he was named emeritus professor in 1988.

I was born on October 2nd 1917, in Thames-Ditton, near London. My parents, of Belgian-German extraction, were Belgian nationals who had taken refuge in England during the war. They returned to Belgium in 1920, and I grew up in the cosmopolitan harbour city of Antwerp, at a time when education in the Flemish part of the country was still half French and half Flemish. Due to these various circumstances, when I entered the Catholic University of Louvain in 1934, I had already travelled in a number of European countries and spoke four languages fairly fluently. This turned out to be a valuable asset in my subsequent career as a scientist.
That I would embrace such a career was, however, very far from my mind. My education, according to the tradition of the jesuit school which I attended, had been centered on the "ancient humanities", and I was strongly attracted to the more literary branches. I nevertheless decided to study medicine, largely because of the appeal of medical practice as an occupation. Medical studies left a fair amount of free time in those days, and there was a tradition at the university that the better students joined a research laboratory. So it was that I entered the physiology laboratory of Professor J. P. Bouckaert, whose rigorous analytical mind exerted a strong influence on my intellectual development. I was attached to a group investigating the effect of insulin on glucose uptake. By the time when I graduated as an MD in 1941, I had abandoned all thought of a medical career, and had only one ambition: to elucidate the mechanism of action of insulin.

Belgian biochemist, born in London, who in recent years has argued that that the evolution of life and mind on the Earth was inevitable. This formed the subject of his 1995 book Vital Dust . de Duve shared the 1974 Nobel Prize for Medicine for his contributions to understanding the structural and functional organization of cells, including his discovery of lysosomes. In 1962 he was appointed a professor at the Rockefeller Institute in New York and has divided his time since between here and the University of Louvain in Brussels, from which he received an M.D. (1941) and the degree of "Agrégé de l'Enseignement Supérieur" (1945) for a thesis on the mechanism of insulin. He is now emeritus professor at both institutions and plans, in his own words, "to devote my remaining years to further probing what, if anything, our growing understanding of life and mind can tell us about the structure and meaning of the universe."

The Leishmaniases
The leishmaniases is a group of diseases caused by protozoan haemoflagelates of the genus Leishmania. These protists belong to the family of the Trypanosomatidae (order Kinetoplastida) and are closely related to the trypanosomes. The disease is tranmitted by female sandflies ( Phlebotomus or Lutzomya ) that feed on the blood of an animal or human host. Leishmaniasis is a zoonosis . It affects mainly sylvatic animals such as rodents and opossums and domestic animals such as dogs. The disease is prevalent in most tropical and sub-tropical areas of the world, such as East and South-East Asia, the Middle Esat, North and East Africa, the south of Europe (the Mediterranian basin) and Central and South America. It is worth bearing in mind that this kind of book passes its sell-by date relatively quickly (it was written in about 1995); new hypotheses are continually being generated and tested, and old ones rejected. That said, 'Vital Dust' is comprehensive in its scope, and I think that anyone with an interest in biology, evolution and the possibility of extraterrestrial life would find much to chew on herein.
