Jean-Baptiste-Gabriel-Joachim Dausset
French hematologist and immunologist whose studies of the genetic basis of the immunological reaction earned him a share (with George Snell and Baruj Benacerraf) of the 1980 Nobel Prize for Physiology or Medicine.
After serving with the Free French forces in World War II, Dausset resumed his interrupted medical studies and took his degree from the University of Paris in 1945. He pursued advanced studies at Harvard University and later became laboratory director of the National Blood Transfusion Centre. From 1958 to 1977 he was associate professor and then professor of immunohematology at the University of Paris, and from 1977 he was professor of experimental medicine at the College de France. In investigating the immunological reactions of patients who had received many blood transfusions, Dausset correctly hypothesized that a specific genetic variation among people accounted for the different levels of reaction. He eventually demonstrated the existence of a gene complex that he designated HLA (human leucocyte antigen) and that proved to be analogous to the H-2 complex in the mouse studied by Snell; both came to be seen as types of the major histocompatability complex (MHC) characteristic of all vertebrates.

jean dausset – Biography. It was there that jean dausset was born, on 19th October1916, and this region has held a strong attachment for him ever since.

His choice of career was almost dictated by that of his father, Henri Dausset, who pioneered Rheumatology in France. His medical studies progressed without incident until the advent of the Second World War, when they were interrupted. He was mobilized in 1939 and returned from the French Campaign in 1940 to a Paris occupied by the Germany Army. He began to devote his time ardently to the preparation of a competitive examination for the title of Intern of the Paris Hospitals. Upon receiving this title, he immediately left to join the fighting forces in North Africa. During the Tunisian Campaign, he performed blood transfusions in the army. This was his first introduction to immunohaematology.
While training in Algiers, he performed his first laboratory experiments and carried out his first scientific study on blood platelets.

As soon as the war was over, he undertook his first real research study, in collaboration with Professor Marcel Bessis. Professor Bessis had just developed exchange-transfusion in new-born babies and adults. It is impossible to say how much time he spent treating, with this method, women who had become anuric following abortion manoeuvres resulting in septicaemia due to Clostridium perfringens - this was his first contact with kidney failure!

Alu-PCR hybridization
Pooling strategy
Alu-PCR products (amplified individually with A33 Alu primer, Chumakov et al, Nature Genetics, 1, 222-225, 1992 ) of 24576 YACs (from 734_a_1 to 989_h_12) were pooled according to a 3-dimensional scheme. This library was divided in 64 superpools of 4 microtiter plates each. Then each superpool (384 clones) was represented by 22 pools:
  • 8 pools of half-plates (the first pool contains all half of the first plate, the second pool contains the second half of the first plate, and so on). Each of these pools contains Alu-PCR products of 48 YACs.
  • 8 pools containing Alu-PCR products of the 64 clones of lanes A to H.
  • 6 pools containing columns 1+7, 2+8, 3+9, ... , 6+12 ( Alu-PCR products of 64 clones each).
The total number of pools to test for screening this library is 1408 (64x22). All of them can be contained in a 16-density filter of the size of a microtiter plate.
Monosomic somatic cell hybrid panel
In order to get information about chromosomal location of each Alu-PCR probe, we added tin the membranes two copies of a monosomic cell hybrid panel. This panel comes mainly from NIGMS mapping panel 2. We spotted on membranes Alu-PCR product of these hybrids.
