A dominant theme of research in my laboratory during the past ten years has been the development and application of recombinant DNA and related methodologies for the analysis of genetic recombination in eukaryotic cells.
An important goal in cell and molecular genetics of mammalian cells is to be able to target exogenous genes to specific loci in the genome. This has been achieved although the frequency is low. Our current program aims to discover more about the process of gene conversion, a non-reciprocal recombination process in which sequences from one duplex DNA replace a corresponding region in a homologous duplex DNA. Both in vitro and in vivo experiments are being pursued to explore this reaction, particularly the isolation of yeast mutants blocked in their ability to carry out gene conversion between episomal and chromosomal homologous sequences. We are exploring this strategy for targeting alterations in chromosomal genes of embryonic stem cells with a goal of creating mutant animals.
The HIV-1 virus induces an immunodeficiency disease in patients it infects by depleting the T-helper population. We are exploring the signal transduction pathway from the T cell receptor as a possible target for the action of one of the viral genes.

US molecular biologist. In 1972, using gene-splicing techniques developed by others, Berg spliced and combined into a single hybrid the DNA from an animal tumour virus (SV40) and the DNA from a bacterial virus. For his work on recombinant DNA, he shared the 1980 Nobel Prize for Chemistry.
Berg was born in New York and educated at Pennsylvania State University and Case Western Reserve University. After graduating from Pennsylvania State College in 1948 and taking a doctorate from Western Reserve University in 1952, Berg pursued further studies at the Institute of Cytophysiology in Copenhagen and at Washington University in St. Louis, where he remained as assistant professor of microbiology until 1959. In 1956 Berg identified an RNA molecule (later known as a transfer RNA) that is specific to the amino acid methionine. He then perfected a method for making bacteria accept genes from other bacteria. This genetic engineering can be extremely useful for creating strains of bacteria to manufacture specific substances, such as interferon. But there are also dangers: a new, highly virulent pathogenic microorganism might accidentally be created, for example. Berg has therefore advocated restrictions on genetic engineering research.
From 1959 he was associated with the medical school of Stanford University, serving as chairman of the biochemistry department in 1969-74 and becoming Willson professor (1970) and director of the Beckman Center for Molecular and Genetic Medicine (1985).

He chairs the USB-IF Device Working Group, and co-chairs (with MCCI's Terry Moore ) the Communication Device Class committee. He has been involved with the development of USB technology since its inception. He recently was technical editor of the USB-IF Logo Compliance Testing test procedure document. Formerly Director of Engineering, USB Products at SystemSoft Corporation, he was directly involved with the development of PCMCIA technologies including CardBus, Zoomed Video, and Multi-Function cards. Paul has chaired several PCMCIA sub-committees; and served as the Technical Editor for the 1995 PC Card Standard. While working with PCMCIA he was a systems architect and software specialist at Ventura Micro, Inc. and at the Zenith Data Systems Advanced Technology Center in Santa Clara.

In the course of studying the actions of isolated genes, Berg evolved methods for splitting DNA molecules at selected sites and attaching segments of the molecule to the DNA of a virus or plasmid, which could then enter bacterial or animal cells. The foreign DNA was incorporated into the host and caused the synthesis of proteins that were not ordinarily found there. One of the earliest practical results of recombinant technology was the development of a strain of bacteria containing the gene for producing the mammalian hormone insulin.
