Volume 81, Number 25 CENEAR 81 25 pp. 53-54, 56 ISSN 0009-2347 MEDICINAL CHEMISTRY Working in the field of drug discovery requires a broad mix of interests and skills LOUISA WRAY DALTON J im Burnett exited from his small liberal arts college, Randolph-Macon, Ashland, Va., with a biology degree and a chemistry minor. "After I graduated, I still didn't really know what I wanted to do," he says. "I knew that drugs were kind of cool, thoughthe legal kindthe ones that effect change in a biological system. I liked the idea of manipulating a living system with a small molecule." So Burnett entered a Ph.D. program in medicinal chemistry at Virginia Commonwealth University. Burnett is one of a growing number of young chemists entering the field of medicinal chemistry. According to an annual survey conducted by the American Chemical Society, as of March 1, 2003, 14.8% of ACS members under age 40 specialize in medicinal or pharmaceutical chemistry, compared with 9.8% in the membership overall. The younger the ACS members are, the more likely they are to be somehow affiliated with making drugs. And medicinal chemistry is ultimately just that: using all the tools of chemistry and biology possible to create a molecule that treats human disease.

BSc (Hons.) Medicinal Chemistry will appeal to students with an interest in chemistry in relation to the medical and health sciences, such as pharmaceutical chemistry, clinical chemistry, and medical laboratory science. Students complete a number of core chemistry and biomedical modules and then specialise in modules relating to medicinal aspects of chemistry. Modules available include Clinical Chemistry, Analytical Chemistry, Modern Synthetic Methods, Molecular Cell Biology, Pharmacology . Some of the specialist medicinal modules are delivered at Addenbrookes Hospital in Cambridge. Aims of the course
The course aims to provide a comprehensive understanding and appreciation of medicinal and chemical sciences by detailed studies of modules selected from the fields of biology, biomedical sciences and chemistry. The department has a long tradition and extensive experience mainly in the field of steroid chemistry and in the analytical characterization of steroid hormones. Starting from basic steroids, extensive and complex projects are conducted here for preparing new compounds for pharmacological investigations and clinical trials. The medicinal chemistry used for this objective includes modern catalytic and organo-metallic reactions, combinations of chemical and enzymatic processes, parallel synthesis as well as computational chemistry. Steroid analysis is very closely connected to steroid chemistry. Direct access to modern analytical methods allows fast structure elucidation and characterization of newly synthesized compounds. The chemists focus on lead finding and optimization. Accordingly, they are involved early in project generation, closely interacting with structural and molecular biologists, pharmacologists and physicians. The most recent results from the Department of Medicinal Chemistry which have found international attention and acknowledgement are linked to new estrogen sulfamates mesoprogestins and steroids with radical scavenging properties which will be further investigated on an inter-disciplinary basis and in co-operation with international partners.

Combinatorial libraries are synthesised for screening against novel targets as part of the Centre's new initiative in combinatorial chemistry/high throughput screening. The Mass Spectrometry Group provides analytical support for combinatorial chemistry and for pharmacokinetic and drug metabolism studies on novel compounds in preclinical and early clinical development. The Team provides medicinal chemistry input into the discovery and development of inhibitors of novel molecular targets. Combinatorial Chemistry During the 18 months since the robotic facility for combinatorial synthesis was installed, we have synthesised two medium sized libraries (~1000 compounds in each). From one of these we have identified lead inhibitors of target enzymes histone acetyl transferase and ERK2. We have also developed resin-based methodologies for the synthesis of several further compound libraries.
