Dr. Li, along with his wife Liu Yingli and daughter Diana, met with Smith to thank him for his efforts on their behalf. Ph.D. University of Birmingham, UK 1959 Molecular Biology of DNA-damage induced mutagenesis. Our work is concerned with Studies with E. coli. carried out in collaboration with Roger Woodgate (NIH) and Myron Goodman (USC), are concerned with understanding the in vivo functions of the UmuD'C protein complex, which plays a crucial role in replication past DNA damage in this organism. In particular, we are attempting to identify the DNA polymerase holoenzyme subunits with which the complex interacts, and to determine the relative roles of DNA polymerase II and III. These questions are being investigated by transforming different mutant strains with single stranded vectors that carry a specified mutagenic lesion at a defined site. Such vectors are powerful tools for this purpose because they provide direct and independent estimates of the frequency of replication past the lesion and of the frequency with which mutations are induced by this process. The laboratory has two main research areas:
(1). To develop a strategy that targets proteins, especially pathogenic proteins, to the proteasome pathway. Immune system detects infections by recognizing antigenic peptides presented by the MHC molecules on the surface of the cell. The proteasome pathway is the main source of peptides for the MHC Class I presentation. The transcription factor NF-?B p50 is generated through a unique cotranslational mechanism by the proteasome, and a glycine-rich region (GRR) is required for such processing. Since sequences downstream of the GRR are not required for recognition by the proteasome, and these sequences might be channeled into the proteasome after the initial cleavage, we plan to fuse different genes to sites downstream of the GRR and test whether these sequences will be targeted for degradation. We will study the theoretical basis of the targeting mechanism, and test whether such targeting can stimulate high production of specific cytotoxic T cells. (2). To Study the mechanism of protein dimerization. Many proteins can form homo- and heterodimers and thus serve diverse biological functions. However, the rules governing this common cellular phenomenon remain unclear. NF-?B is a group of proteins that dimerize through their Rel homology domain (RHD) to form homo- or heterodimers. Among them, NF-?B 1 proteins can homodimerize during translation, and such dimerization may serve as a necessary step for the subsequent processing mediated by the proteasome. We plan to use NF-?B as a model system to study the dimerization mechanism. We are interested in studying both the primary structure of a protein that determines or partitions its homo- and heterodimerization, and the cellular factors that mediate such processes. Biochemical, biophysical and molecular biology approaches will be used for the study.

Email C.J.H.Duggan@reading.ac.uk His books include Fascism and the Mafia (Yale University Press, 1989) and A Concise History of Italy (Cambridge University Press, 1994). He has recently completed a major biography of the 19th-century Italian statesman, Francesco Crispi, published in Italy, and subsequently in Britain as Francesco Crispi. From nation to nationalism (Oxford University Press, 2002 ). He is currently working on aspects of nation-building in 19th- and 20th-century Italy. He is also Director of the Centre for Modern Italian History
