ORTALDO@NCIFCRF.GOV The LEI consists of three sections: Experimental Therapeutics Section (ETS) Cellular and Molecular Immunology Section (CMIS) , and Leukocyte Cell Biology Section (LCBS) The LEI has focused and coordinated approach toward the understanding the innate immune system and how it can interact with adaptive immunity, leading to the development of therapeutic strategies for the treatment of cancer. The rapid, systemic response of an organism to malignant cells and pathogens in their environment is known as the innate immune response. It is characterized by synthesis and/or release of immunomodulatory molecules and activation of cellular processes that directly or indirectly attack the pathogen. In addition to innate immunity's role in combating malignant cells and pathogens, increasing evidence has emerged related to a pivotal role for the innate immune system in driving and shaping adaptive immunity. The Leukocyte Cell Biology Section of the LEI has been focused on the discovery and characterization of a class of novel mouse NK receptors has led to the identification of Leukocyte Regulatory Receptors [LRR] that are involved in positive and negative regulation of immune cell function. Direct evidence has now been demonstrated that NK cells and expression of their newly discovered array of Ly-49 NK receptors is required for the adaptive immune response to xenografts. The mechanisms by which grafted cells interact with the innate immune system is an important target for future investigation and potential therapeutic. The molecular basis of lymphocyte cytotoxic effector functions and their importance in the immunotherapy of cancer is being pursued by the laboratory of Dr. T. Sayers.

El Centro de Inmunología Molecular tiene como principal misión obtener y producir nuevos biofármacos destinados al tratamiento del cáncer y otras enfermedades crónicas no transmisibles e introducirlos en la Salud Pública cubana. Hacer la actividad científica y productiva económicamente sostenible y realizar aportes importantes a la economía del país. El 5 de Diciembre de 1994 en el oeste de la Habana se inaugura el Centro de Inmunología Molecular. Para el diseño y construcción de este centro se utilizaron las actuales guías para las buenas prácticas de producción adoptadas por Cuba y recomendadas por la Organización Mundial de la Salud. Particularmente aquellas adoptadas también por los países miembros de la Comunidad Europea. La edificación es básicamente una construcción biplanta que abarca más de 15 000 metros cuadrados.

The Department of Microbiology and Immunology is focussed on research and educational programs which relate to microbial pathogens (viruses and prokaryotic and eukaryotic microbes) and the host immune defenses which protect against these organisms. Current research emphasizes cutting-edge molecular techniques and includes, but is not limited to, studies on: autoimmunity, biodefense, bioinformatics biofilms, cancer biology, gene therapy, genetics, HIV/AIDS, immunologic mechanisms and vaccine development. The University of Rochester has long been considered among the top 25 research universities in the United States. Recent evaluations of the Department of Microbiology and Immunology, including rankings by national peer review groups and others place our Department among the very best in the country. Furthermore, NIH grant support in the Dept. of Microbiology and Immunology has grown considerably in the past five years, rising by almost 300%, from $4.4 million in FY 1999 to $12.0 million in FY 2003. Experimental Immunology Section
Research
The interest of the Experimental Immunology Section is to learn about ocular antigens, the immune responses they provoke, the inflammatory diseases induced by these responses and the mechanisms that modulate these diseases. Specific studies under current investigation include: 1) The mechanisms of immunotolerance against ocular antigens are being investigated , focusing mainly on the role of the thymus in this process. These studies include: (i) Cells expressing ocular-specific antigens are being identified in thymi of wild type mice, using immunohistochemical methods. (ii) Apoptosis of lymphocytes specific toward ocular antigens is being actually visualized in thymic sections of double transgenic mice expressing a foreign antigen (hen egg lysozyme, HEL) in their lens and HEL-specific T-cell receptor (TCR) on their T-cells. The apoptosis effect is demonstrated by reduced HEL specific responsiveness in the periphery of these mice. 2) Cellular mechanisms of ocular inflammation are being investigated, using transgenic mice expressing HEL in their lens as recipients for T-cells expressing HEL-specific TCR. Purified subsets of lymphocytes (Th0, Th1, Th2) are being prepared and their features are being compared with regard to the their pathogenicity and the typical histopathological changes they provoke in the recipient mice.
