Mucin stains
There are a variety of mucin stains, all attempting to demonstrate one or more types of mucopolysaccharide substances in tissues. The types of mucopolysaccharides are as follows:
  • Neutral - These can be found in glands of the GI tract and in prostate. They stain with PAS but not with Alcian blue, colloidal iron, mucicarmine, or metachromatic dyes. Acid (simple, or non-sulfated) - Are the typical mucins of epithelial cells containing sialic acid. They stain with PAS, Alcin blue at pH 2.5, colloidal iron, and metachromatic dyes. They resist hyaluronidase digestion. Acid (simple, mesenchymal) - These contain hyaluronic acid and are found in tissue stroma. They do not stain with PAS, but do stain with Alcian blue at pH 2.5, colloidal iron, and metachromatic dyes. They digest with hyaluronic acid. They can be found in sarcomas. Acid (complex, or sulfated, epithelial) - These are found in adenocarcinomas. PAS is usually positive. Alcian blue is positive at pH 1, and colloidal iron, mucicarmine, and metachromatic stains are also positive. They resist digestion with hyaluronidase. Acid (complex, connective tissue) - Found in tissue stroma, cartilage, and bone and include substances such as chondroitin sulfate or keratan sulfate. They are PAS negative but do stain selectively with Alcian blue at pH 0.5.

This section contain histology laboratory protocols which are in daily use in our laboratories in Nottingham. At present there is an index of about 100 techniques which we will expand as time allows. Nottingham is the home of the textbook Theory and Practice of Histological Techniques edited by J.D. Bancroft and A. Stevens. Churchill Livingstone. The fourth edition has just been published and was launched in 1996. We would like to invite contributions to this web page and try to expand it. Ed's Basic Histology Gallery This site was placed primarily for the medical students at The University of Health Sciences in Kansas City. It introduces basic concepts of histology actively, without having to schedule classroom time. You will need basic histology competency in the years ahead.
  • You will need it to make sense of what's happening at the tissue level in "Pathology", in reading professional journals, and when you are caring for sick people. You will need it to complete your three "Pathology" signoffs and hundred-or-so lab cases successfully. You will be expected to review at least some of your own biopsies with the pathologist, regardless of your speciality.
Students who complete this exercise can expect to be successful on the first histology signoff. (We are considering this as a requirement for completing the first unit of the new curriculum.) In this exercise, you will be required to identify, all by yourself, under a real microscope with a flesh-and-blood instructor, EACH of the following:

The vagina is a canal that extends from the external vulva to the cervix . The vaginal walls can be easily distended but are normally 7.5 cm in length anteriorly and 9 cm long posteriorly. The upper portion of the vagina encircles the vaginal part of the uterine cervix. The vagina touches the empty bladder on the ventral and superior surface. Inferiorly the vagina adheres to the posterior wall of the urethra and opens adjacent to the labia minora. The vaginal lining has many transverse ridges, or folds. Vaginal tissues receive blood supply from uterine arteries and sometimes branches of the internal iliac artery. Blood returns to the venous system through veins that empty into the internal iliac veins. Lymphatic drainage is via the external and internal iliac lymph nodes and superficial inguinal lymph nodes.
