Molecular Microbiology and Immunology integrates many disciplines (biochemistry, virology, parasitology, molecular biology, immunology,ecology structural biology) concerned with the study of the transmission, immunobiology, and pathogenesis of bacterial, parasitic, viral, and immunological diseases of public health importance. Research is at the population, organismal, cellular, and molecular levels. The central premise of this broad approach to the study of disease is that public health problems can best be addressed by understanding basic biological mechanisms. Our research aims to advance the understanding of the biology of disease and to use this knowledge to solve public health problems. Research takes place in the laboratory, in the clinic, and in the field, as the faculty works to combat such enormous public health problems as malaria, sexually transmitted diseases, mosquito-borne encephalitis, tuberculosis, diarrhea, measles, AIDS, and autoimmune diseases. Department of Biological Sciences, Lehigh University, Bethlehem PA 18015 In this course, we will study viruses at the levels of their molecular biology and genetics, and their interactions with the host organism. Our coverage will focus almost entirely on viruses that infect humans and cause serious disease. The course will be somewhat self-contained on this website. The book Principles of Molecular Virology (3'rd ed., 2001) by A. J. Cann will be a source of some figures. We will also make extensive use of other web sources. Useful sites include All the Virology on the WWW (for background), the U.S. Centers for Disease Control and Prevention (including its journal Emerging Infectious Diseases and its Morbidity and Mortality Weekly Report ) and the World Health Organization . Lehigh has electronic subscriptions to many journals publishing new virology research, including Science the Nature journals (particularly Nature Medicine , the Proceedings of the National Academy of Sciences (PNAS) Virology Journal of General Virology Journal of Virology , and Journal of Infectious Diseases
Grading will be based on three required 100 point exams and a 100 point "new journal articles-web site presentation" project in April, for a total of 400 points. Those of you taking the course as 397 (Virology and Writing) will be graded on the same scale as for 353, except that you will also write a major term paper (an in-depth summary and analysis of some topic in current virology research) to satisfy the "writing intensive" requirement and the extra one credit. There is no final exam.

Research Interests
Very little is known as to how mRNA is exported from the nucleus into the cytoplasm. We are using fission yeast Schiz. pombe as a model system to identify genes that are involved in regulating nuclear export of mRNA. We have identified a temperature sensitive mutant in Schiz. pombe rae1-1 that accumulated poly(A)+ RNA in the nucleus at high temperature. This mutant also conferred a cell cycle arrest at the G2/M boundary at high temperature. However, the inability to export mRNA at high temperature is not restricted to a particular phase of the cell cycle. We have isolated a number of extracopy and extragenic suppressors of mutation . These are being pursued so that we can identify genes that are required for nuclear transport of mRNA and also, pursue those whose function is required for cell cycle progression. The study of these suppressors should allow us to understand if a link exists between mRNA export and cell cycle progression.

Research activities
Research in the Department of Virology deals with plant diseases caused by viruses, phytoplasma and viroids. Studying viral genome structure and function, development and adaptation of diagnostic systems, disease transmission and epidemiology and to development of means to reduce virus damage in protected or field grown crops.
Detection and Identification of Plant Viruses.
Several different technologies have been developed and are currently in use in our Department. These methods include electron microscopy (EM), biological tests, serological and molecular techniques such as PCR, RT-PCR, dsRNA analysis and nucleic acid hybridizations. Most of these methods are being used for the production of virus-free propagation material as well as in commercial laboratories for virus diagnosis in vegetatively-propagated crops (bulbs, corms and stems), vegetables, fruit trees and grapes.
