Microbes include higher and lower organisms, although Oral Microbiology concerns mainly bacteria, some viruses and few fungi:
Structure and Morphology of Microorganisms
Viruses Viruses are not cells, and have no cell membrane or cytoplasm. Viral particles (virions) consist of nucleic acid surrounded by a protective protein coat, although considerable variety is seen in their morphology and structure. Their genetic material may be DNA or RNA, but one viral type will only contain one type of nucleic acid, providing one of the major differential characteristics used in virology. Viruses do not possess the machinery for synthesising macromolecules, and so are completely dependent on a host cell for their replication. Upon entry of viral nucleic acid into a host cell, the viral components are synthesised within the host cell, viral particles are assembled and finally released by lysis or budding from the host cell membrane. Viruses are not sensitive to antibiotics. Prokaryotes and Eukaryotes There are fundamental differences in eukaryotic and prokaryotic cell structure and gene expression; the defining difference is the presence of a nuclear membrane surrounding the genetic material of eukaryotes, but not prokaryotes.

Evolutionary origin of land plants; biology of charophycean algae and basal bryophytes; microbial associations with freshwater algae and peat mosses blank I try to understand evolutionary transitions related to origin and early diversification of land plants, which likely occurred more than 500 million years ago. My students and I use phylogenies, together with molecular, physiological, ultrastructural, paleontological, and ecological methods, to trace the first appearance of fundamental plant features such as embryo, cuticle, walled spores, meristems, plastid carbonic anhydrases, and bacterial and fungal associations. This work necessarily focuses on charophycean green algaethe modern protists most closely related to plants, and bryophytesthe earliest-divergent plant lineages. Recent research students have worked on : 1) molecular ecology of eubacteria associated with peat mosses ( Sphagnum spp.) from northern Wisconsin bogs, with an emphasis on methane oxidizers, 2) molecular ecology of eubacteria associated with charophycean green algae of northern Wisconsin bogs, with an emphasis on nitrogen fixers, 3) evolution of meristems, plasmodesmata, and cuticles, 4) Ordovician fossils from the Madison area, including bryophyte-like remains and the earliest-known fossil fungi (Glomales, a group known to commonly form symbiotic associations with plants), 5) hexose transporter genes and proteins of charophycean algae and bryophytes, 6) TEM immunolocalization studies and activity measurements of carbonic anhydrases in charophycean algae.

Evidence suggests that mechanisms involved in the initiation and establishment of bacteria in surface communities (biofilms) may also operate in the development of bacterial/host associations. Further, it is often a judgement call (and dependent on environmental factors) whether an association is classified as either symbiotic or pathogenic. For these reasons, I am interested in both biofilm formation and pathogenic microbiology for clues they may provide to understanding marine bacterial/invertebrate interactions. My current research focus is the association of marine a-proteobacteria with the Eastern oyster, Crassostrea virginica , and specific goals are to understand: (i) the transition of the bacteria from a free-living state to association with host tissues, (ii) the role of cell-cell signaling (e.g. quorum sensing), (iii) factors controlling the specificity of the associations, and (iv) host defense responses.

Course Description This course is an introduction to microorganisms, small animal forms, and their activities and roles in terrestrial ecosystems, as well as various plant-microbial associations. A prior course in microbiology is not necessary, but chemistry through one semester of organic is very desirable. The material is divided into four major sections:
  • Basic material on soil organisms and techniques for studying them in soil. Concepts of microbial distribution and function as influenced by soil characteristics and environmental factors are described. Elemental cycles of carbon, nitrogen, sulfur, and phosphorus. The emphasis of this section is on common pathways used by microbes for degradation, transformation, and synthesis of compounds containing these elements and on applications to soil productivity and environmental issues. Degradation of xenobiotic compounds. A vast array of synthetic organic compounds (xenobiotics) are used in industry or are deliberately applied to the soil for control of insects, diseases, and weeds. Land application is also a suitable method for disposal of many agricultural and some industrial wastes. Principal degradation pathways and environmental fate of the major groups will be considered.
