    CYTOKINES
    The term cytokine includes the lymphokines and monokines (terms which are actually still used in the literature but nevertheless confusing). Complex interactions among various cells of the immune response are mediated by a group of secreted low-molecular weight proteins that are collectively designated as cytokines. Cytokines are the messenger molecules of the immune system. Generally, they act locally , not systemically.
    General Properties:
    Secreted by one cell type act upon cells which bear a receptor for the cytokine. Very high affinity between the cytokine and its receptor which allows even very small quantities of cytokines to have a potent effect. When the cytokine binds to its receptor, it triggers intracellular signals (signal transduction pathways) which lead to specific changes in gene expression. autocrine - binding to the same cell that secreted it
    paracrine - binding to a nearby cell
    endocrine - binding to a distant cell
    Actions of cytokines may be:
    pleiotropic (they elicit different biological activities from different target cells.

    Mature B lymphocytes bear ~150,000 immunoglobulin molecules on their surface. Each of these 150,000 immunoglobulins( Ig ) possess an identical binding site for antigen. In other words, a particular B lymphocyte is specific for only one antigen. In addition to surface Ig, B lymphocytes possess other characteristic surface proteins:
    Class II MHC proteins (MHC stands for Major Histocompatability Complex)
    The presence of Class II MHC proteins permits B lymphocytes to act as antigen presenting cells ( APCs ) to T helper lymphocytes.

    T he Division of Transplantation Pathology is an integral part of the comprehensive transplant program at the University of Pittsburgh Medical Center . Its mission is to provide up-to-date laboratory services emphasizing continued excellence in patient care, to foster an academic environment devoted to education and training, and to conduct basic and clinically oriented research in transplantation pathology.     LaboratoryServices
    T he Division offers a wide range of laboratory tests and tissue analyses for patients and for experimental studies. Routine histology, immunocytochemistry and in situ hybridization are performed on tissues and fine needle aspirates from patients under consideration for transplantation, native organ resections and post-transplant biopsies. These procedures assure consistency of analysis and continuity of patient care. Post-transplant monitoring assays deal with lymphocyte reactivity and cytokine production and on detection of cytomegalovirus, Epstein-Barr virus, and hepatitis viruses. Procedures for fine needle aspiration analysis have been added to post-transplant immunologic monitoring.

    Building on the existing strong programs in Immunology, Microbiology and Vaccine Biology at the University of Rochester, the Center for Vaccine Biology and Immunology focusses on basic research into immunological mechanisms, and also on the long-term goal of helping to design the next generation of vaccines. The Center occupies 31,500 square feet of research space on the top floor of the recently (1999) constructed Kornberg Medical Research Building at the University of Rochester Medical Center. In addition, faculty throughout the Medical Center, including those in the new research building, share state-of-the-art core facilities which are housed in the new research building and elsewhere.
