Future outbreaks may be hard to predict, tests suggest.
16 January 2004 HELEN R. PILCHER Infected gorilla carcasses can spread Ebola to people who touch them. A genetic analysis of Ebola in central Africa has helped confirm that the disease can spread to people when hunters handle infecetd animal corpses. Eric Leroy at the International Centre for Medical Research in Franceville, Gabon, and colleagues, along with local villagers, noticed an increased number of large animal carcasses in the forest just before one Ebola outbreak in Gabon. When the researchers tested the corpses, most were found to be positive for the Ebola virus. The team then looked at the genetic sequences of viruses in both animal and human victims from five outbreaks that struck Gabon and the Republic of Congo over the past two years . They traced each outbreak from dead animals through to the first human victim and subsequent human victims. Their analysis suggests that the five outbreaks were caused by five distinct Ebola viral strains, rather than by one strain that mutated into different forms. That implies that a new outbreak could pop up anywhere at any time, says Leroy, making future outbreaks very difficult, if not impossible, to predict.

7 August 2003 This story is from the news section of the journal Nature Ebola outbreaks occur every few years in Africa. High-speed vaccines could help to contain unexpected outbreaks of disease, say US researchers who have developed a fast-acting vaccine against the deadly Ebola virus. The new vaccine, reported in this issue of Nature , gives macaque monkeys protection from Ebola infection only four weeks after a single jab. Previous Ebola vaccines took six months and multiple boosters to confer immunity. Should the vaccine prove safe and effective in humans, this would be fast enough to stop Ebola outbreaks, says lead author Gary Nabel, head of the Vaccine Research Center at the National Institute of Allergy and Infectious Diseases in Bethesda, Maryland. It could also form a template for developing emergency vaccines against unexpected outbreaks of disease, Nabel says. Most vaccines serve only as a preventive measure, because they take too long to produce immunity to be useful once an outbreak has begun. A notable exception is smallpox vaccine, which acts so quickly that it can sometimes stop the disease even after a person has been infected with the virus.

WEB EXCLUSIVE Bill Delaney: Ebola scare in Canada HAMILTON, Ontario (CNN) Health officials in Canada said Wednesday preliminary tests on a woman feared to have the Ebola virus have been negative for the deadly hemorrhagic fever. However, they said viral isolation tests are still being conducted and they have not yet been able to rule out other viruses, such as Lassa or yellow fever, as the cause of her illness. "We're being extremely cautious," said Dr. Mark Loeb of Henderson Hospital, where the woman is being treated. But he emphasized the general public is not at risk. "We've had some results from the laboratory in Winnipeg, showing that the Ebola testing is negative," explained Loeb. "The real implication (is) that this greatly reduces the likelihood that this patient has been infected with the Ebola virus." The disease is caused by infection with Ebola virus, named after a river in the Democratic Republic of the Congo (formerly Zaire) in Africa, where it was first recognized. The virus is one of two members of a family of RNA viruses called the Filoviridae. Three of the four species of Ebola virus identified so far have caused disease in humans: Ebola-Zaire, Ebola-Sudan, and Ebola-Ivory Coast. The fourth, Ebola-Reston, has caused disease in nonhuman primates, but not in humans.
Scientists and researchers are faced with the challenges of developing additional diagnostic tools to assist in early diagnosis of the disease and ecological investigations of Ebola virus and the disease it causes.

WASHINGTON (CNN) A civilian Army researcher at Fort Detrick, Maryland, is in isolation after possibly being exposed to the Ebola virus, Army officials said Thursday. The researcher accidentally pricked herself with a needle that contained a weakened form of the Ebola virus last week while she was injecting mice with the virus as part of a research effort. The woman has shown no signs of the fatal illness, but will remain at Fort Detrick for up to 30 days of isolation. Local government officials have been notified, but no one else is believed to have been exposed.

The leader of the team that designed an Ebola vaccine and tested it in macaque monkeys expects to see no adverse effects in the human trial The subjects will be monitored for 1 year, but Nabel anticipates no adverse effects, because other DNA vaccines have already proven safe. Plus, the vaccine contains no infectious materials from Ebola virus. Instead, he told The Scientist The current trial is just the first test of a vaccine that Nabel's group described 3 years ago in a report to Nature , detailing the first successful test of an Ebola vaccine in a nonhuman primate. That trial employed a prime/boost strategy. Once a month for 3 months, macaque monkeys were injected with plasmid DNA encoding Ebola virus glycoprotein and nucleoprotein to prime the immune system. Three months later (5 months after the first DNA injection), the monkeys received an injection of crippled adenovirus containing the glycoprotein gene to boost the immune response. The team then challenged the vaccinated animals 3 months later with a lethal dose of Ebola virus (at 8 months). Significantly, the vaccine induced both humoral and cellular immune responses in the animals. For instance, recognizing that the prime/boost strategy's multipart regimen could prove impractical, Nabel's team in August described an accelerated vaccine in another
