Boston, MA- A team of researchers from the Harvard School of Public Health, led by Gökhan Hotamisligil of the Department of Nutrition and collaborators Lufen Chang and Michael Karin from University of California, San Diego, have identified the gene JNK (c-Jun Amino-terminal kinases) as the key component in interfering with insulin sensitivity in the metabolic pathway for obesity, obesity-induced insulin resistance and type 2 diabetes. The discovery identifies a new target for therapeutic drugs for both obesity and diabetes. The research findings appear in the November 21 issue of the journal Nature Obesity is closely associated with insulin resistance and is one of the leading risk factors for type 2 diabetes, both affect more than 50 percent of the US population. Little has been known about the molecular mechanisms between these two metabolic diseases. Both are associated with a wide range of inflammatory molecular activity in fatty tissue. These actions activate the JNK that interfere with insulin sensitivity. Type 2 diabetes usually occurs after age 40. People with this type of diabetes do not produce adequate amounts of insulin for the needs of the body and/or cannot use insulin effectively. To test the role of JNK (also known to researchers as "Junk") in decreasing insulin sensitivity in a variety of obesity models, the team bred mice lacking either form of the gene JNK1or JNK2 and mice possessing the JNK genes and fed both groups a high fat, high calorie diet to induce obesity. A control group of lean mice with identical genotypes was fed a standard diet. JNK activity and protein concentrations were highest in liver, muscle and fat tissue in mice with the JNK1 gene that were on the high fat diet compared to lean control mice and mice fed a high fat diet that lacked the JNK gene. Weight gain rose the sharpest over an eight-week span for the mice on the high fat diet but particularly for those with the JNK genes. In studying total body fat composition, JNK-deficient mice had significantly decreased total body fat accumulation compared to the other groups.

Diabetes Prevention: The Test
Diabetes is about to become a concern for millions of middle-aged Americans who never gave it much thought before. The American Diabetes Association, backed by federal health authorities, recently called for routine screening of all Americans starting at age 45, using a simple, inexpensive test, in order to detect more cases of Type 2 diabetes earlier. This form of diabetes, previously called non-insulin dependent or adult-onset diabetes, accounts for 90 to 95% of cases of diabetes. Until now, the disease has generally been diagnosed when people develop symptoms, such as unusual thirst or frequent urination, or when people at high risk for the disease (such as those with a family history of it) are tested for it. Now blood sugar testing may become as routine as blood pressure screening.
Diabetes is a breakdown in the body's ability to utilize glucose (blood sugar) efficiently. Glucose, the main sugar into which foods are digested, can be used by our cells only in the presence of the hormone insulin. With Type 2 diabetes, the cells become resistant to the effects of insulin, and thus blood levels of glucose rise. (With Type 1, which is usually diagnosed in young people, the body virtually stops producing insulin.) The incidence of Type 2 diabetes is on the rise, largely because the U.S. population is aging and getting heavier.

If you have a small frame, subtract ten percent.     By TINA ALTAMIRANO Waging battle in the weight-loss war, the recent low carbohydrate diet craze exhibits promise as a possible treatment for diabetes. As the seventh leading cause of death for Hispanics, diabetes is a perpetual concern in the Latino community.
    Often linked to weight control and food selection, diabetes ailments include foot maladies, blindness, and kidney and heart diseases that often result from the excessive and erratic blood sugar, or glucose levels that characterize the disease. Prescribed by an increasing number of doctors in conjunction with medication to control and stabilize diabetics' dangerous glucose levels, low carbohydrate diets eliminate or severely restrict many foods that directly affect these levels. Steve Hopkins, coordinator for Walk the Cure, a research fundraiser at the Juvenile Diabetes Foundation in Austin, Texas, says that his low carbohydrate diet plan "allows a lot more bang for your food buck.
