Powerful molecular, genetic and physical techniques have been developed to analyze living systems at increasingly higher levels of resolution, down to the level of genes and molecules. This has led toan enormous increase in our knowledge of basic processes in biology. The faculty in the Department of Molecular, Cell and Developmental Biology use molecular biology and genetics to study multicellular model organisms such as mouse, zebrafish, Drosophila and Arabidopsis . Our goal is to understand processes such as cell signaling, gene regulation, circadian rhythms, and morphogenesis. The molecular genetic approach will be particularly important in the years to come, as a means of deciphering the function of the myriad of genes indentified in the human genome, as well as that of model organisms. Extractions: OVERVIEW: Our research is aimed at understanding the molecular mechanisms which regulate neuronal injury and survival following acute or chronic CNS insults. Our emphasis is on those circuits neural which govern motor behavior. Following many forms of neural injury, damaged neurons are eliminated through an autonomous process known as programmed cell death (PCD). Aberrant regulation of PCD has been shown to be an important component of the neural degeneration observed in disorders such as Amyotrophic lateral sclerosis, Alzheimer's disease, Parkinson's disease and Huntington's chorea; as well as acute injury states such as spinal cord injury and stroke. Understanding the biological mechanisms which control PCD is thus a key element in enhancing our ability to promote neuronal survival and functional recovery following CNS injury. To examine the processes of PCD and neural re-innervation, our laboratory utilizes techniques in molecular biology, biochemistry, genetics, ex vivo cell culture, and microsurgery. Through the targeted modification of mammalian genes, anatomic and morphometric analyses of the murine CNS, primary neural cell culture, and neural transplantation, we are examining the molecular activation and regulation of downstream PCD effectors during neural injury
