Extractions: snowhill@library.ucsb.edu The University of California's California Digital Library (CDL) formed an Ebook Task Force in August 2000 to evaluate academic libraries' experiences with electronic books (e-books), investigate the e-book market, and develop operating guidelines, principles and potential strategies for further exploration of the use of e-books at the University of California (UC). This article, based on the findings and recommendations of the Task Force Report ], briefly summarizes task force findings, and outlines issues and recommendations for making e-books viable over the long term in the academic environment, based on the long-term goals of building strong research collections and providing high level services and collections to its users. While electronic books and texts have been available for some time for selected public domain titles, only relatively recently have electronic texts been packaged and offered commercially as electronic books. It is the shift to the commercial production, sale, and distribution of e-books that has changed how libraries need to deal with e-books, and what prompted our investigation.

If the poison primer binding site is removed by a deletion, a full length PCR product can be enriched, allowing second round amplification of the deleted PCR product. We tested the poison primer method on a 133 bp deletion in the gene dim-1, using two different internal poison primers (A and B) that lie within the deletion interval (Figure 2). One or both were included in the external round of nested PCR, with different mixtures of wild type and deletion mutant DNAs used as the beginning template at the indicated ratios.With no poison primer, both the 701 bp wild type PCR fragment and 568 bp deletion fragment were observed in reactions with WT:Mutant ratios up to 10:1, but the ability to detect the deletion was lost at higher ratios. With the poison primer, we could detect the deletion fragment at ratios of 5000:1.
